Background The inhibition of quorum sensing (QS) is considered as a novel strategy for development of antipathogenic agents and management of nosocomial infections caused by Pseudomonas aeruginosa. The present study aimed to investigate the anti-quorum sensing activity of sub-MIC concentrations of quercetin (Q), imipenem (IPM) and ciprofloxacin (CIP) alone and in combination against P. aeruginosa. Methods We determined the MIC and synergistic activity of Q, IPM and CIP against P. aeruginosa PAO1. The effect of sub-MIC (1/4 and 1/16 MIC) concentrations of Q, IPM and CIP alone and in combination on QS related virulence factors and expression of lasI and lasR genes was also determined. Results The addition of Q decreased the MIC of IPM and CIP and there was additive effect from these combinations. Quercetin did not show significant effects on cell growth at concentrations of 1/4 and 1/16 MIC. However, sub-MIC concentrations (1/4 and 1/16 MIC) of Q in combination with IPM or CIP showed the significant reduction in QS dependent phenotypes (P < 0.001). The combination of Q with IPM or CIP at the concentrations of 1/4 MIC showed the greatest reduction in the expression levels of lasI compared with untreated control (P < 0.001). The docking results suggested that quercetin has potential to inhibit the LasR protein. Conclusion Considering the beneficial biological effects of quercetin and its inhibitory effects on QS related virulence traits, the combination of this natural compound with imipenem or ciprofloxacin may be used as an effective approach for managing of multidrug resistant infections of P. aeruginosa.

Bacteria can migrate in groups of flagella‐driven cells over semisolid surfaces. This coordinated form of motility is called swarming behavior. Swarming is associated with enhanced virulence and antibiotic resistance of various human pathogens and may be considered as favorable adaptation to the diverse challenges that microbes face in rapidly changing environments. Consequently, the differentiation of motile swarmer cells is tightly regulated and involves multi‐layered signaling networks. Controlling swarming behavior is of major interest for the development of novel anti‐infective strategies. In addition, compounds that block swarming represent important tools for more detailed insights into the molecular mechanisms of the coordination of bacterial population behavior. Over the past decades, there has been major progress in the discovery of small‐molecule modulators and mechanisms that allow selective inhibition of swarming behavior. Herein, an overview of the achievements in the field and future directions and challenges will be presented.

In primary ciliary dyskinesia (PCD) patients, Pseudomonas aeruginosa is a major opportunistic pathogen, frequently involved in chronic infections of the lower airways. Infections by this bacterial species correlates with a worsening clinical prognosis and recalcitrance to currently available therapeutics. The antimicrobial peptide, lin-SB056-1, in combination with the cation chelator ethylenediaminetetraacetic acid (EDTA), was previously demonstrated to be bactericidal against P. aeruginosa in an artificial sputum medium. The purpose of this study was to validate the anti-P. aeruginosa activity of such a combination in PCD sputum and to evaluate the in vitro anti-virulence effects of EDTA. In combination with EDTA, lin-SB056-1 was able to significantly reduce the load of endogenous P. aeruginosa ex vivo in the sputum of PCD patients. In addition, EDTA markedly reduced the production of relevant bacterial virulence factors (e.g., pyocyanin, proteases, LasA) in vitro by two representative mucoid strains of P. aeruginosa isolated from the sputum of PCD patients. These results indicate that the lin-SB056-1/EDTA combination may exert a dual antimicrobial and anti-virulence action against P. aeruginosa, suggesting a therapeutic potential against chronic airway infections sustained by this bacterium.
