Inhibitory Activities ofCudrania tricuspidataLeaves on Pancreatic LipaseIn Vitroand LipolysisIn Vivo

Abstract: To identify effective herb to treat obesity, we screened 115 herbal extracts for inhibition of porcine pancreatic lipase (triacylg-ycerol acylhydrolase, EC 3.1.1.3) activity in vitro. Of the extracts tested, Cudrania tricuspidata leaves exhibited the most pronounced inhibitory effect on lipase activity with an IC50 value of 9.91 μg/mL. Antilipid absorption effects of C. tricuspidata leaves were examined in rats after oral administration of lipid emulsions containing 50 or 250 mg  C. tricuspidata/kg body weight… Show more

“…Orlistat, which reduces dietary fat absorption by inhibiting pancreatic lipase, is currently on the global market [ 4 ]. Pancreatic lipase is a major enzyme involved in triglyceride absorption in the intestine, and inhibiting fat absorption from the diet is a target for treating obesity [ 5 ]. However, these drugs have many limitations due to adverse effects, such as stomach pain, steatorrhea, and headaches [ 6 ].…”

“…One such plant is Cudrania tricuspidata (CT), which is used in Korean folk medicine ( 25 – 27 ). One study reported that CT leaves inhibited lipase activity, reduced plasma triacylglycerol levels, and delayed lipid absorption in vivo ( 28 ). In addition, recent studies found that CT has anti-inflammation and antihyperglycemia effects ( 29 31 ).…”

“…Previous human clinical trials have shown that lipase inhibitors from plants can effectively reduce fat absorption to suppress weight gain in obese patients [12][13][14][15] , the findings in this work are not only very helpful to well understand the molecular interactions between pancreatic lipase and polyphenols extracts from peanut shells, but also greatly contribute to the development of the natural effective inhibitors for the prevention of human obesity in future. Therefore, polyphenols extracts from peanut shells may be useful in treatment of obesity.…”
