Inhibitory Effects of BiRyuChe-Bang on Mast Cell-Mediated Allergic Reactions and Inflammatory Cytokines Production

Abstract: BiRyuChe-bang (BRC) is a Korean prescription medicine, which has been used to treat allergic rhinitis at Kyung Hee Medical Center. In this work, we investigated the effects of BRC on mast cell-mediated allergic reactions and inflammatory cytokines production, and identified the active component of BRC. Histamine release was measured from rat peritoneal mast cells (RPMCs). Ear swelling and passive cutaneous anaphylaxis (PCA) were examined in mouse models. Phorbol 12-myristate 13-acetate (PMA) plus A23187-induce… Show more

“…). Most of the screenings for TNF‐α inhibitors reported in literature have been carried out with one single concentration, in the range from 10 to 1000 µg/mL (Chou et al, ; Moon et al, ; Kim et al, ; Zhong et al, ). Therefore, the data here reported demonstrate unequivocally that the active extracts promote a concentration‐dependent inhibition of this important pro‐inflammatory cytokine.…”

“…The extract concentration employed in the present work was defined based on literature records for similar screenings. A wide assortment of concentrations is reported, ranging from 10 to 1000 μg/mL (Chou et al, 2013;Moon et al, 2013;Kim et al, 2013;Zhong et al, 2013). Some crude extracts elicit marginal TNF-α inhibition when tested at low concentrations, but the maximum effect does not reach high values (Wangchuk et al, 2013).…”

“…Besides that, BRC suppresses histamine, mRNA expressions of TNFalpha, IL-6, and IL8, and the activation of NFkappaB in a human mast cell line (HMC-1). Therefore, this formulation can be used to treat mast cell-mediated allergic and inflammatory responses 53 . According to the evidence, the medicinal plants, their derivatives, and several herbal formulations in oriental medicine are used to treat allergic rhinitis and, via different action mechanisms, lead to decrease in the symptoms or even treatment of this disease.…”

“…In addition, serum levels of eosinophile cationic protein 55 and OVA-specific IgG1 56 , several interleukins, biogenic aminos, and interferongamma were reported to decrease after administration with medicinal plants or their compounds 44,53 . Besides that, anti-allergy medicinal plants affect chloride channel-3 and suppress secretion of MCP-1 and CIC-3, which play an important role in chemo attraction of the leukocytes to the sites of allergic responses and especially pathological process of allergic rhinitis, as well as VCAM-1 in the epithelial mucus and therefore controls inflammatory responses 56 .…”

“…The RNA was quantified by measuring the absorbance at 260 nm. The RNA (1 g) from each sample was reverse transcribed to cDNA with PrimeScript reverse transcriptase (TaKaRa Co, Ltd., Shiga, Japan) and oligo (dT) primer (Promega Co, Madison, WI, USA) (Moon et al, 2013). To amplify the selected protein transcripts, the following gene primers (Invitrogen, Carlsbad, CA, USA) were used: ERK1 5 0 -GCT GAA TCA CAT CCT GGG TAT-3 0 (forward) and 5 0 -AGA TCT GTA TCC TGG CTG GAA-3 0 (reverse); ERK2 5 0 -GCA GGT GTT CGA CGT GGG AAT-3 0 (forward) and 5 0 -GTG CAG AAC ATT AGG TGA ATA-3 0 (reverse); yielding predicted products of 373 and 394 bp, respectively.…”
