Inhibitory effects of openers of large-conductance Ca2+-activated K+ channels on agonist-induced phasic contractions in rabbit and mouse bronchial smooth muscle

Abstract: Airway smooth muscle expresses abundant BKCa channels, but their role in regulating contractions remains controversial. This study examines the effects of two potent BKCa channel openers on agonist-induced phasic contractions in rabbit and mouse bronchi. First, we demonstrated the ability of 10 μM GoSlo-SR5-130 to activate BKCa channels in inside-out patches from rabbit bronchial myocytes, where it shifted the activation V1/2 by −88 ± 11 mV (100 nM Ca2+, n = 7). In mouse airway smooth muscle cells, GoSlo-SR5-1… Show more

“…The efficacy of some of them has been reported to depend on the presence of BK channel regulatory β and γ‐subunits (Kshatri et al, ; Large et al, ; Webb et al, ). These compounds, in particular GoSlo‐SR‐5‐130 and GoSlo‐SR‐5‐6,activated BK channels in freshly isolated smooth muscle cells from rabbit bladder (Large et al, ), rabbit corpus cavernosum (Hannigan et al, ), and bronchial smooth muscle (Bradley et al, ). Furthermore, Webb et al () demonstrated that the effects of GoSlo‐SR‐5‐6 were reduced by >80%, when a triplet of mutations were introduced on the S4/S5 linker and S6 helix.…”

“…42) But it has also been reported that Ca 2+ signaling can relax artery smooth muscle and ASMCs by activation of BK channels. 43,44,45) In ASMCs, Ca 2+ signaling induced by the activation of bitter taste receptors opens BK channels and thus induces membrane hyperpolarization and the relaxation of ASMCs. 21) Besides the opening of BK channels, the inhibition of voltage-dependent L-type Ca 2+ channels and non-selective cation channels have also been suggested to be involved in TAS2Rs-triggered relaxation of ASMCs.…”
