Injectable hydrogel as a carrier of vancomycin and a cathelicidin‐derived peptide for osteomyelitis treatment

Abstract: A local drug delivery system that attempts to find a suitable balance between antimicrobial and regenerative actions was developed for osteomyelitis treatment (OM). This system combines the angiogenic and immunomodulatory peptide LLKKK18 (LL18) and vancomycin hydrochloride (VH), loaded into an injectable oxidized dextrin (ODEX)‐based hydrogel (HG). In vitro cytotoxicity was analyzed in MC3T3‐E1 pre‐osteoblasts and erythrocytes. The kinetics of LL18 release was studied. Antimicrobial activity was assessed in vi… Show more

“…Besides, to provide an improved antibacterial capability for in vivo applications, this NC hydrogel system could be advanced by using black titanium dioxide with a better sonocatalytic property, 104,105 incorporating catechol groups with a tissue adhesion property, 106,107 and conjugating antibacterial segments (e.g., antibacterial peptide 108,109 or polyethyleneimine 110 ) to provide a sustained antibacterial effect.…”

“…In a clinical MRSA‐induced osteomyelitis model in mice, LL18 exerted an immunomodulatory effect in a dose‐dependent manner as compared to the use of 28 m m VH, and a concentration of 300 µ m combined with 483 µ m VH eradicated the infection in 70% of individuals and tissue damage (Figure 11C). [ 133 ] Osteomyelitis treatment has encouraged further research into the ability of AMPs to clear infection while stimulating bone regeneration. Overall, AMPs can be combined with other biocompatible, cost‐effective, and easy‐to‐manufacture biomaterials such as HG as a noninvasive delivery system for osteomyelitis treatment, which is a promising strategy.…”
