Integrated Karyotypes of Diploid and Tetraploid Carrizo Citrange (Citrus sinensis L. Osbeck × Poncirus trifoliata L. Raf.) as Determined by Sequential Multicolor Fluorescence in situ Hybridization With Tandemly Repeated DNA Sequences

Abstract: Deng et al. Karyotypes of Carrizo Citrange Rootstock the integrated karyotype and chromosomal characteristics of this important citrus rootstock as well as its spontaneously occurring tetraploid plant. Furthermore, this study is a good prospective model for study species with morphologically indistinguishable small chromosomes.

“…(AG 3 T 3 ) 3 , a classic chromosome end marker, is typically located in the distal region of the chromosome in H. mutabilis [ 32 ], J. regia , J. sigillata [ 31 ], F. pennsylvanica , S. oblata , L. lucidum , L. × vicaryi [ 28 ], B. diaphana , and B. soulieana [ 28 ]. Other similar types of (T x A y G z ) n [ 49 ] have also been identified at each chromosome end in the woody plants C. sinensis × P. trifoliata [ 23 ], Citrus clementina Hort. Ex Tan.…”

“…Typical chromosomal loci largely used as FISH probes are represented by tandem repetitive arrays of 5S and 45S rRNA genes and satellite DNA sequences whose chromosome location is species-specific (e.g., [25,[39][40][41][42]). Another extensively employed probe types are BAC (bacterial artificial chromosome) clones bearing large single-and low-copy DNA sequences [43,44] or certain mobile repetitive elements dispersed on all chromosomes but provide signals on specific chromosomal regions, e.g., in centromeres [8,[45][46][47].…”
