INTERACTION BETWEEN PTERIDINE SYNTHESIS AND RIBOFLAVIN ACCUMULATION INDROSOPHILA MELANOGASTER

Abstract: Using chromatographic and spectrophotometric methods, riboflavin was determined to be the major component of yellow pigment in the Malpighian tubes of wild-type Drosophila melanogaster. While reduced yellow pigment accumulation in larvae of eye color mutants is consistently correlated with a reduction of drosopterin formation in adults, reduction of drosopterin synthesis is not always associated with decreased yellow pigment in Malpighian tubes. In the light mutant, reduced yellow pigment accumulation per se is… Show more

“…Riboflavin is an essential B vitamin in nearly all pro- and eukaryotic organisms, serving as a precursor for the synthesis of flavin coenzymes and flavin adenine dinucleotide which are essential cofactors for a wide variety of metabolic enzymes and electron transport38. In addition to metabolic processes, riboflavin is also involved in areas of yellow pigmentation in the mosquito, such as within the eyes and malpighian tubules39. The accumulation of riboflavin in mosquitoes exposed to live M. ulcerans , and the reduced accumulation in mosquitoes exposed dead M. ulcerans is surprising (Figure 5).…”

“…Here we describe a LRO in the Malpighian tubules of Drosophila that forms the major physiological zinc storage site in this animal. This zinc storage granule concentrates the entire chelatable pool of total body zinc in flies, and is distinct from the previously described riboflavin-containing granules that give the wild type tubule its characteristic yellow-orange color (NICKLA 1972).…”

“…Moreover, another common feature of all LRO-biogenesis mutants is a reduction of riboflavin containing pigment granules in their Malpighian tubules (BREHME and DEMEREC 1942;NICKLA 1972). Generally, yellow Malpighian tubules correlated with normal zinc content, whereas loss of coloration correlated with low body zinc ( Figure 4).…”

“…The emergence of highthroughput technologies has given us possibilities to systematically survey and study the underlying biological system. The molecular interaction maps have been built in model organisms (e.g., S.cerevisiae [8], D.melanogaster [9] and C.elegans [10]), as well as in higher vertebrate organisms (e.g., zebrafish [11], mouse [12] and human [13]). Evidently, the generated interaction maps offer us a rich resource for systematic studies of molecular networks and complement other types of biological data.…”
