Interaction of Genetic Variations inNFE2L2andSELENOSModulates the Risk of Hashimoto's Thyroiditis

Abstract: Background: Several single-nucleotide polymorphisms (SNPs) are known to increase the risk of Hashimoto's thyroiditis (HT); such SNPs reside in thyroid-specific genes or in genes related to autoimmunity, inflammation, and/or cellular defense to stress. The transcription factor Nrf2, encoded by NFE2L2, is a master regulator of the cellular antioxidant response. This study aimed to evaluate the impact of genetic variation in NFE2L2 on the risk of developing HT. Methods: In a case-control candidate gene associatio… Show more

“…Strong evidence indicates that epigenetics plays a significant role in regulating oxidative stress. Nuclear factor erythroid 2-related factor 2 (NRF2), encoded by nfe2l2 (7), is one of the major regulators of oxidative stress (8). Numerous studies have explored the key role of NRF2-related epigenetic modifications in various disease models (9,10).…”

“…Selenoprotein S, a member of the selenoprotein family, is expressed in multiple tissues and is involved in cellular stress responses and immune and inflammatory processes (Santos et al, 2019). In this study, knockdown of SeS by siRNA partially reversed E. coli-induced increases in TLR4, IKB, NF-κB, TNF-α, and IL-1β.…”

“…The important role of genetics in this context can be recognized in recent studies by Santos et al [195] , demonstrating a relation between the SELENOS promoter genotype and NFE2L2 promoter genotype, which together modulate the risk of Hashimoto's thyroiditis. SELENOS is of importance since it constitutes a basic element of the endoplasmic reticulum, especially under conditions of glycolytic metabolism [196] .…”
