Intercellular calcium waves integrate hormonal control of glucose output in the intact liver

Abstract: Key points Sympathetic outflow and circulating glucogenic hormones both regulate liver function by increasing cytosolic calcium, although how these calcium signals are integrated at the tissue level is currently unknown. We show that stimulation of hepatic nerve fibres or perfusing the liver with physiological concentrations of vasopressin only will evoke localized cytosolic calcium oscillations and modest increases in hepatic glucose production. The… Show more

“…The cytosolic concentration of calcium in normal resting cells is very low (about 0.1μM) and is more than 10,000 times lower than its concentration in blood plasma (about 2 mM). Numerous extracellular signals from hormones to growth factors are transduced to intracellular [Ca 2+ ]i spikes that are amplitude and frequency encoded [73][74][75]. In addition, they are highly localized within cells [76].…”

“…Под влиянием ДХИ за счет снижения липидного обмена и поглощения свободных жирных кислот в гепатоцитах подавляется печеночный глюконеогенез. В результате повышается чувствительность тканей к инсулину [27]. Кроме того, инозитолы участвуют в секреции инсулина в поджелудочной железе.…”

“…In insulin-sensitive tissues, the posttranslational activation of glucokinase (one of the rate-limiting enzymes of glycolysis) requires an increase in cytoplasmic Ca 2+ levels (Markwardt et al, 2016). Variations in Ca 2+ oscillations also regulate the generation of glucose from non-carbohydrate precursors (gluconeogenesis) (Gaspers et al, 2019) by modulating the expression of genes related to glucose metabolism in the liver (Wang et al, 2012;Zou et al, 2018). Furthermore, in response to Ca 2+ -mobilizing hormones, Ca 2+ controls the phosphorylation state, and hence the activity, of key enzymes regulating glucose storage (glycogen) (Amaya and Nathanson, 2013).…”
