Interference of Xenoestrogen o,p′-DDT on the Action of Endogenous Estrogens at Environmentally Realistic Concentrations

Abstract: Estrogenic effects of individual chemicals and mixtures of 17β-estradiol (E2) and o,p'-DDT were examined using pS2 and PR gene expressions in MCF-7 cells. Interactions between E2 and o,p'-DDT were evaluated by a statistical method based on factorial design. At levels close to environmentally realistic concentrations, no significant interactions between E2 and o,p'-DDT were observed, suggesting that the xenoestrogen (o,p'-DDT) could act in an additive way with the endogenous estrogen (E2). These results demonst… Show more

“…This means the combinations with endogenous estrogens and environmental estrogens occur in additive way and contribute to the total estrogenicity present in human plasma. With a statistical method, mixtures effects of E 2 and o,p'-DDT were examined using pS2 and PR gene expressions in MCF-7 cells [34]. The results suggested that the xenoestrogen (o,p'-DDT) could act in an additive way with the E 2 , even at levels close to environmentally realistic concentrations [34].…”

Nevertheless, considering the differences between in vitro and in vivo situation, the concentration in the environment or encountered by humans can not be fully compared with the medium concentration.…”

“…Knowing that estrogen mediates pro-reproductive effects through actions in several organs of the female reproductive system (ovaries, fallopian tubes, uterus and placenta formation) and biochemical processes (Rosselli et al 2000;Deroo & Korach 2006;Zoloaga et al 2013), experimental studies of reproduction regarding the adverse effects caused by frequent exposure to xenoestrogen substances have been performed, reaffirming the importance and constant level of exposure at which the current world population is exposed to (Bromer et al 2010;Zhong et al 2013). Thus, the present study tested the hypothesis that melatonin can block or reduce the estrogenic effects of cimetidine in uterine stroma, interfering with ERα-receptor expression, content of collagen fibers and hormone levels (estrogen, progesterone and prolactin) in adult rats.…”

“…From in vivo studies, o,p'-DDT has been found to be estrogenic, as measured by uterotrophic responses in mammals and vitellogenins production in fish [1,6]. O,p'-DDT has been identified as a relatively potent estrogen agonist in in vitro tests with estrogen-responsive tumor cells and in the yeast estrogen screening (YES) assay [2,7].…”
