Interferon-Mediated Response to Human Metapneumovirus Infection

Abstract: Human metapneumovirus (HMPV) is one of the leading causes of respiratory diseases in infants and children worldwide. Although this pathogen infects mainly young children, elderly and immunocompromised people can be also seriously affected. To date, there is no commercial vaccine available against it. Upon HMPV infection, the host innate arm of defense produces interferons (IFNs), which are critical for limiting HMPV replication. In this review, we offer an updated landscape of the HMPV mediated-IFN response in… Show more

“…The response elicited by the innate immune system upon hMPV infection plays an essential role in the primary control of the disease and the subsequent activation of the adaptive immune response [ 47 , 48 , 49 , 50 , 51 , 52 ]. In the following section, the innate immune response elicited during infections with hMPV will be discussed as well as how the innate immune cells activate the interferon pathways.…”

“…The interferon response, particularly that of the respiratory epithelium, is so critical for hMPV restriction that the virus has acquired ways to halt IFN-I secretion [ 47 ]. In this line, the SH protein of hMPV is capable of inhibiting STAT1 phosphorylation ( Table 1 ) and thus inhibiting the transcription of many ISGs [ 67 , 68 ].…”

“…In addition, the G protein has been reported to interact with RIG-I and to block RIG-I-dependent gene transcription and thus IFN production [70]. For a detailed overview of the interferon response upon hMPV infection in vitro, animal models and clinical settings, we refer to [71].…”

“…During natural infection, HMPV employs different strategies to circumvent the host’s interferon response to the pathogen, thus resulting in impaired innate and adaptive immune responses [ 51 ]. HMPV modulates host immunity by inhibiting the activation of cytokine signaling; this mechanism depends on the cell type, and there is some variation in virus strains.…”
