Interleukin‐10 mRNA expression in B‐cell chronic lymphocytic leukaemia inversely correlates with progression of disease

Abstract: Interleukin-10 (IL-10) has been shown in vitro to inhibit survival and spontaneous DNA synthesis in B-cell chronic lymphocytic leukaemia (B-CELL) cells by induction of programmed cell death. We have analysed the presence of mRNA transcripts for IL-10 in purified B-CLL cells from 35 patients by RT-PCR. Transcripts for IL-10 were detected in 11/20 patients with non-progressive disease. In cell preparations from patients with progressive B-CLL IL-10 mRNA were detected in only 2/15 samples (P < or = 0.01). The Eps… Show more

“…Many of these lymphomas have been shown to be comprised of CD5ϩ B cells (24,25). We as well as others have reported that IL-10 mRNA levels can also be varied in CLL (6,8,26). However, the NZB mouse, the murine model of CLL, has consistently elevated serum IL-10 levels and increased IL-10 mRNA expression in the malignant B-1 cells.…”

“…Complementary DNA (cDNA) was prepared from the RNA using reverse transcriptase (RT) polymerase chain reaction (PCR) analysis. The total mRNA was assessed by performing a PCR analysis with the primers for b 2 -microglobulin [16], which is structurally expressed by all cells. The mRNA for a given cytokine in the individual cell type cDNA preparation was determined in the PCR using primers designed by Pisa et al for IFNg [17] and IL-12 (Pharmacia Biotech, Uppsala, Sweden) and IL-10 (Scandinavia Gene Synthesis, Kö ping, Sweden).…”

“…Lymphoma. Tumor cells from B, T, and NK cell lymphoma are able to produce biologically active IL-10 ( Kitabayashi et al, 1995;Masood et al, 1995;Sjoberg et al, 1996;Beatty et al, 1997;Boulland et al, 1998;Jones et al, 1999). As early as 1993, Favrot and coauthors (Blay et al, 1993) investigated IL-10 serum levels using an ELISA, which detects both viral and human IL-10 in patients with active non-Hodgkin's lymphoma (NHL) and healthy volunteers.…”
