Interleukin-12 Production by Human Alveolar Macrophages Is Controlled by the Autocrine Production of Interleukin-10

Abstract: By releasing interleukin (IL)-12 in the lung, alveolar macrophages (AM) may profoundly modify an immune response. The autocrine regulation of the heterodimeric, biologically active form of IL-12 (IL-12 p70) by IL-10 was studied, as well as the expression of its subunits of 35 kD (p35) and 40 kD (p40). AM cultured in medium alone expressed only p35 mRNA. Both p35 and p40 mRNA levels were induced by lipopolysaccharide (LPS) and were further increased by interferon-gamma (IFN-gamma). LPS alone induced IL-12 p40 b… Show more

“…From this, we estimate the relative effect parameter f 5 ϭ 4.8. Incorporation of results from several other similar studies (58,59,111,112) extend this estimate of f 5 to the range (4.8, 65). Other saturation parameters.…”

“…Nevertheless, IgA had no significant effect on MAPK activation that could underlie its inhibitory effect on IL-12. We previously showed that IgA may induce IL-10 expression in monocytes and MD-DCs (15), and IL-10 may also control IL-12 expression in an autocrine manner (16,36), as observed following IgG ligation of FcgRs. However, autocrine IL-10 was not involved in the IgA-mediated inhibition of IL-12.…”

“…IL-12p70, composed of p35 and p40 subunits, is a major Th1-driving cytokine establishing cell-mediated immunity (6) and preferentially expressed by APC cells such as pulmonary macrophages (21,22). We have therefore studied the relative contribution of this cytokine and its subunits in the development of a mouse model of pulmonary fibrosis induced by silica particles.…”
