Intermolecular crosslinking and stereospecific molecular packing in type I collagen fibrils of the periodontal ligament

Abstract: A trypsin digest of denatured NaB3H4-reduced native bovine periodontal ligament was prepared and fractionated by gel filtration and cellulose ion-exchange column chromatography. Prior to trypsin digestion, a complete acid hydrolysate was subjected to analyses for nonreducible stable and reducible intermolecular cross-links. Minute amounts of the former and significant amounts of the reduced cross-links dihydroxylysinonorleucine (1.1 mol/mol of collagen), hydroxylysinonorleucine (0.9 mol/mol of collagen), and h… Show more

“…6). However, biochemical studies have shown that the periodontal ligament exhibits no significant changes in the quantities of the major reducible collagen cross-links during development and maturation (Pearson et al, 1975), and shows only small amounts of mature, nonreducible cross-links (Yamauchi et al, 1986) in the bovine incisor. Further studies are needed to clarify the discrepancy between polarized light microscopic and biochemical studies.…”

“…The specific activity of the NaB 3 H 4 was determined by the method described previously (34,35). The reduced samples were washed with cold distilled water several times by repeated centrifugation at 4,000 ϫ g and lyophilized.…”

“…The glycosylated immature reducible cross-links were analyzed employing base hydrolysis of the reduced samples as described above (34). By applying the hydrolysates to the HPLC system, the glycosylated (GG-and G-) and non-glycosylated cross-links were separated.…”

“…Collagen cross-linking sites reportedly correlate with the molecular packing of collagen fibrils and subsequent nucleation of BAp [67]. Although the bio-mineralization process has been thought to be directed by acidic non-collagenous proteins, such as small integrin-binding ligand N-linked glycoprotein [68], it has been reported that type I collagen can initiate bio-mineralization in the absence of any other extracellular matrix molecules in vitro [69].…”
