“…A few of the important pathways are visualized here. OCM is also important for providing cofactors and substrates for a multitude of subsequent reactions ranging from epigenetic methylation of genes to purine production folate, and riboflavin (Garrod et al 2010;Herbert and Zalusky 1962;Silla et al 2011;Yamada et al 2001;Yang et al 2008). “…MTR is also responsible for the catabolism of 5-methyltetrahydrofolate to tetrahydrofolate. Low MTR activity leads to a buildup of folate in the 5-methyltetrahydrofolate form, termed the ''methyl folate trap'' (Herbert and Zalusky 1962). MTRR, depicted in Fig.…”

“…Only MG forms crossed the enterocyte's barrier as confirmed by several works in animals 30,31 , in humans 14,29,32,33 and in vitro 15,34 . The folate conjugase responsible for this hydrolysis, also called pteroyl polyglutamates hydrolase or polyglutamate hydrolase, has been detected in many tissues and medias: intestine 35,36 , blood, bile, pancreas, kidney [37][38] , brain 31 and liver 30 . Within the enterocytes, the Tetrahydrofolic Acid (THF) is produced from dihydrofolic acid by folate reductases and then converted into 5 methyl tetrahydrofolates which is the only form joining the portal circulation 19,31,39 ( Figure 2).…”

“…Also by 1962, Herbert and Zalusky, at the Thorndike, first proposed the 'folate trap hypothesis', defining the interrelationships of vitamn B 12 and folic acid. They showed that folic acid activity 'piles up' in human serum in the presence of B 12 deficiency and that this accumulation of N5 methyl-tetrahydrofolic acid provides direct evidence of deranged folic acid metabolism due to vitamin B 12 deficiency (Herbert & Zalusky, 1962). Rosenberg et al (1969) demonstrated that intestinal mucosal enzymes participate in absorption of folate from peptide conjugates that predominate in dietary folate.…”
