Intracerebroventricular injection of citrate inhibits hypothalamic AMPK and modulates feeding behavior and peripheral insulin signaling

Abstract: We hypothesized that citrate might modulate the AMP-activated protein kinase/acetyl-CoA carboxylase (AMPK)/(ACC) pathway and participate in neuronal feeding control and glucose homeostasis. To address this issue, we injected citrate into the lateral ventricle of rats. Intracerebroventricular (ICV) injection of citrate diminished the phosphorylation of hypothalamic AMPK/ACC, increased the expression of anorexigenic neuropeptide (pro-opiomelanocortin and corticotropinreleasing hormone), elevated the level of mal… Show more

“…After a 1-week recovery period, cannula placement was confirmed by a positive drinking response elicited by the administration of angiotensin II (2 l of 10 Ϫ6 M solution) (17); rats that drank less than 5 ml of water within 15 min after treatment were excluded from the studies. Adequately cannulated rats were randomly selected for the experimental groups.…”

“…Of note, rather than feeding, thyroid hormones control brown adipose tissue thermogenesis through specific action on AMPK in the VMH [24]. In addition to hormonal signals and neuropeptides, hypothalamic AMPK and ACC are also modulated by nutrients and intermediate metabolites, including glucose [19,25,26], α-lipoic acid [27], citrate [28] and lactate [29]. …”

“…Taken together, these effects may promote increased generation of malonyl CoA. Indeed, citrate increases hypothalamic malonyl CoA content , secondary to decreasing the phosphorylation and activation of AMPK, which subsequently prevents the phosphorylation and inhibition of ACC Stoppa et al, 2008). The anorexigenic effects of citrate are also associated with decreased hypothalamic NPY mRNA expression, but increased POMC mRNA expression .…”
