Intraoperative and post operative treatment with 5-Fluorouracil and mitomycin-c: long term effects in vivo on subconjunctival and scleral fibroblasts

Abstract: Rabbits undergoing full thickness glaucoma filtering surgery were exposed to one of 4 treatments. Group 1 received intraoperative distilled water, group 2 received intraoperative mitomycin-c 0.2 mg/ml for 5 minutes, group 3 received 5 post operative injections of 5-Fluorouracil (5-FU) 5 mg in 0.1 ml, and group 4 received intraoperative 5-FU 50 mg/ml followed by 5 post operative injections of 5-FU. 30 days after the operation tissue biopsies were taken from the subconjunctival and scleral tissue at the treated … Show more

“…On the other hand, after filtration surgery, proliferations of Tenon capsule fibroblasts cause fibrosis of the bleb and surrounding tissue; decreasing the efficacy of the surgery. Intra-operative application of MMC in the center of the deep sclerectomy dissection had been proposed in order to enhance the passage of aqueous from the anterior chamber to the subconjunctival space and to lower the risk of scleral fibrosis (10,(17)(18)(19)(20). This proposal was proven once again in our study.…”

“…First mechanism is about wound healing. Khaw et al showed that MMC stops fibroblast growth for 30 days after surgery (17). Second mechanism is changes in structure and decrease in number of subconjonctival tissue cells.…”

“…The conjunctiva shows an aggressive wound-healing response, and as a result the rabbit has been used extensively in glaucoma research [2,27,38,50,52,53,57,70,107]. It is also an established model in proliferative vitreoretinopathy research [13,24,25,27,48,52,53,62,86,112].…”
