Intravenous feeding of the rat with short chain fatty acid esters II. Monoacetoacetin

Abstract: Acetoacetic acid is oxidized independent of carnitine transport into the cell mitochondria and its monoglyceride is a water soluble compound. The latter was examined as an intravenous nutrient. The monoglyceride of acetoacetic acid was prepared from the acid anhydride, diketene, and glycerol and was found to be totally miscible with water. The nutritional properties of monoacetoacetin were investigated by continuous intravenous infusion of 25 or 50 g/kg body weight per day into ad libitum fed rats. The respons… Show more

“…A monoglycerol ester of β-ketopentanoate is a suitable sodium-free ester, which is rapidly hydrolysed by plasma esterases, like the analogous glycerol monoacetoacetate (Birkhahn and Border 1978). Glycerol mono-β-ketopentanoate, which is much easier to synthesize that the (R)-β-hydroxypentanoate ester, could be used for the acute treatment of fatty acid oxidation disorders.…”

“…MA was prepared from a 1:1 molar ratio of glyc erol and diketene as described by Birkham and Border [4] and tested for both chemical and microbiological purity before and after passage through Millipore (litre and autoclaving. Analysis of the MA was done by thin-layer chromatography.…”

“…DISCUSSION MA is a monoglyceride ester of acetoacetate that was first reported by Birkhahn and Border in 1978. 18 Both acetoacetate and glycerol occur naturally for energy source in mammalian systems, including humans. The molecular weight of MA is 176, its energy is 4.4 kcal/g, and its level of dissolution to water is infinity.…”
