Introgression the Salinity Tolerance QTLs Saltol into AS996, the Elite Rice Variety of Vietnam

Abstract: This study focus on developing new salinity tolerance and high yielding rice lines, using markers assisted backrossing (MABC). Total of 500 SSR markers on 12 rice chromosomes were screened for parental polymorphic markers. Of which, 52 primers in the Saltol region were checked with the two parents varieties to identify polymorphic primers for screening the Saltol region of the breeding populations. For each backcross generation of ASS996/FL478, approx. 500 plants were screened with 63 polymorphic markers distr… Show more

“…The result showed that 28 markers were polymorphic, including 3 markers AP3206, RM3412 and RM10793 on chromosome 1 which linked to Saltol QTL. This result was agreed with some previous reports to verify the polymorphic markers between the parents in target region of Saltol (23)(24)(25) . All those markers were used for screening the generations of the BC1F1, BC2F1 and BC3F1, respectively.…”

“…After three generations of screening with 28 markers, combined with conventional breeding, the promising lines Q5DB-Saltol were selected to develop the next generations. Previously, we also have successfully genotyped BC generations by using the above markers of the crossed of the AS996, BT7, the elite Vietnamese rice cultivars (recipient plants) and FL478 (carrying Saltol QTL, as the donor plant) (6,24) 3,4,5,6,10,11,12,13,16,17,18,19,29,30,32 33,34,36,37,39, 44,45,46 showed homozygous DNA band from Q5DB. Other plants were possessed heterozygous DNA bands…”

“…In this study, the percentage of polymorphic marker was similar with study reported by Linh et al (2012) between BT7 and FL478. Monomorphic primer also were found in this study as also reported in study by Huyen et al (2012) in a study for salt tolerance in rice between (AS996 and IR50404). Monomorphic markers were not valuable for selection process and discarded from marker screening.…”

“…Marker assisted backcrossing (MABC) is another useful method for introgression or substitution of a target gene/QTL from salt tolerant parent to recipient by applying molecular markers. There are several successful examples where Saltol QTL has been precisely introgressed into elite rice varieties through MABC such as PB1121 and PB6 [107], AS996 [108], Bac Thom 7 [109,110] and BRRI Dhan-49 [111].…”
