Investigations on effects of titanium dioxide (TiO2) nanoparticle in combination with UV radiation on breast and skin cancer cells

Abstract: In this study, we have investigated the chemotherapeutic potential of titanium dioxide (TiO 2 ) nanoparticles on skin and breast cancer cells. The cells have treated with a 75 µg/ml concentration of titanium dioxide and then exposed to UV-A radiation. The combined effects of titanium dioxide and UV-A radiation on cell viability, cell cycle, plasma membrane, mitochondrial membrane potentials and apoptotic activity of the cells were investigated. As a result of the research, an increase was observed in the viabi… Show more

“…Cancer cells often overexpress integrin receptors (αvβ3) on their cell surface [ 18 ]. Targeted delivery of TiO 2 NPs, which have shown cytotoxicity against melanoma cells, conjugated to a Arg-Gly-Asp (RGD) motif, would thus make PDT more selective, promoting binding with integrin αvβ3 [ 18 , 19 ]. TiO 2 NPs, conjugated to RGD, exhibit a cytotoxic effect in αvβ3 integrin-expressing mice melanoma cells but not in the normal cells lacking this integrin [ 18 ].…”

“…To evaluate whether the examined gD-derived peptides can interfere with the BTLA-HVEM interaction, we established an in vitro co-culture model of fully immunocompetent healthy T cells and melanoma cell line. SK-MEL-30 melanoma cell line is used as a model cell line in a wide range of cancer-related studies ( 56 , 57 ). Its high expression of both BTLA and HVEM was reported ( 58 ) and confirmed empirically (data not shown).…”

“…Cell viability was determined using the MTT assay method ( Bilkan et al, 2022 ). Briefly, MTT (5 mg/mL, 20 μL/per well) was added to the cells and incubated at 37°C for 4 h. Subsequently, the solutions were removed, DMSO (100 μL/well) was added to the wells, and absorbance was measured at 490 nm using a microplate reader (BioTek, USA).…”
