Involvement of Endoplasmic Reticulum Stress in Uremic Cardiomyopathy: Protective Effects of Tauroursodeoxycholic Acid

Abstract: Background/Aims: Uremic cardiomyopathy (UCM) is a complication in chronic kidney disease. We investigated if endoplasmic reticulum stress (ERS) is involved in UCM, and determined the efficacy of tauroursodeoxycholic acid (TUDCA) in UCM prevention. Methods: Mice were divided randomly into three groups: sham (saline, i.p), 5/6 nephrectomized (Nx) (saline, i.p) and Nx+TUDCA (250 mg/kg/day, i.p.). Renal function was assessed by measuring serum creatinine, blood urea nitrogen and by periodic acid-Schiff reagent sta… Show more

“…As a commonly used inhibitor of ER stress [45-47], TUDCA has been reported to exert preventive effects on ER stress during the early stage of apoptosis before mitochondrial involvement [44, 48]. In the present study, administration of the ER stress inhibitor TUDCA reduced the expression of the ER stress markers GRP78, p-PERK, and CHOP and attenuated UA-induced apoptosis in H9c2 cells, suggesting that ER stress participates in UA-induced apoptosis.…”

“…ERS has been shown to contribute to Aldo-induced renal injury, while treatment with ERS inhibitors (e.g. chemical ER chaperones like tauroursodeoxycholic acid) attenuate Aldo-induced renal injury and uremic cardiomyopathy [16, 30]. …”

“…Simian virus 40 (SV40) and certain strains of Escherichia coli are endocytosed via receptors resident in caveolae. Besides, Cav-1 can help deliver Chl from the endoplasmic reticulum (ER) to caveolae on plasma membrane [10, 39, 40]. At this juncture, Chl can either be transferred to extracellular lipoproteins or be siphoned into the bulk plasma [41-44].…”
