The aim of this study was to determine the effect of equine chorionic gonadotropin (eCG) applied in postpartum (pp) period on ovarian follicle development, serum estradiol concentration, serum progesterone concentration, the calving-to-first oestrus (calving-to-first insemination) interval, and conception at first insemination. The material of this study consisted of 20 cows on day 14 pp. Cows included in this study were randomly divided into two groups. Cows in the first group, were treated with eCG (500 IU Folligon; Intervet, Holland) for 14 days. No treatment was applied to the second group, and it was left as a control group, Ovarian follicle diameters were measured by ultrasonographic examination on days 14, 16, 18, 20 and 22 pp in all animals included in the study. Serum estradiol and progesterone were from blood samples taken on the same days. All animals included in the study were followed up until the first observable oestrus was detected, and they were inseminated 12 hours after the appearance of oestrus symptoms. A pregnancy examination was performed on the 45th day after insemination. In cows administered with eCG during postpartum period, an increase in ovarian follicle diameters compared to untreated cows and a decrease in the calving-to-first oestrus interval were determined. In contrast, serum estradiol and progesterone hormone levels did not differ between the two groups. Pregnancy rate in the experimental group was higher than that in the control group, but there was no statistical difference between them. As a result, it was concluded that eCG applied to dairy cows on day 14 pp did not affect serum estradiol and progesterone levels, but it affected the ovarian follicle diameter and the calving-to-first oestrus interval positively.

Recombinant technologies have made possible the production of a broad catalogue of proteins of interest, including those used for animal production. The most widely studied proteins for the animal sector are those with an important role in reproduction, feed efficiency, and health. Nowadays, mammalian cells and fungi are the preferred choice for recombinant production of hormones for reproductive purposes and fibrolytic enzymes to enhance animal performance, respectively. However, the development of low-cost products is a priority, particularly in livestock. The study of cell factories such as yeast and bacteria has notably increased in the last decades to make the new developed reproductive hormones and fibrolytic enzymes a real alternative to the marketed ones. Important efforts have also been invested to developing new recombinant strategies for prevention and therapy, including passive immunization and modulation of the immune system. This offers the possibility to reduce the use of antibiotics by controlling physiological processes and improve the efficacy of preventing infections. Thus, nowadays different recombinant fibrolytic enzymes, hormones, and therapeutic molecules with optimized properties have been successfully produced through cost-effective processes using microbial cell factories. However, despite the important achievements for reducing protein production expenses, alternative strategies to further reduce these costs are still required. In this context, it is necessary to make a giant leap towards the use of novel strategies, such as nanotechnology, that combined with recombinant technology would make recombinant molecules affordable for animal industry.

Current purification of the glycoprotein equine chorionic gonadotropin (eCG) from horse serum includes consecutive precipitation steps beginning with metaphosphoric acid pH fractionation, two ethanol precipitation steps, and dialysis followed by a numerous of fixed-bed chromatography steps up to the specific activity required. A promising procedure for a more economic purification procedure represents a simplified precipitation process requiring only onethird of the solvent, followed by the usage of magnetic ion exchange adsorbents employed together with a newly designed 'rotor-stator' type High Gradient Magnetic Fishing (HGMF) system for large-scale application, currently up to 100 g of magnetic adsorbents. Initially, the separation process design was optimized for binding and elution conditions for the target protein in mL scale. Subsequently, the magnetic filter for particle separation was characterized. Based on these results, a purification process for eCG was designed consisting of (i) pretreatment of the horse serum; (ii) binding of the target protein to magnetic ion exchange adsorbents in a batch reactor; (iii) recovery of loaded functionalized adsorbents from the pretreated solution using HGMF; (iv) washing of loaded adsorbents to remove unbound proteins; (v) elution of the target protein. Finally, the complete HGMF process was automated and conducted with either multiple single-cycles or multicycle operation of four sequential cycles, using batches of pretreated serum of up to 20 L. eCG purification with yields of approximately 53% from single HGMF cycles and up to 80% from multicycle experiments were reached, with purification and concentration factors of around 2,500 and 6.7, respectively.
