Involvement of Phytophthora spp. in chestnut decline in the Black Sea region of Turkey

Abstract: Summary Chestnut blight caused by Cryphonectria parasitica is a serious disease of Castanea sativa in the Black Sea region of Turkey. During disease surveys, dieback and decline symptoms were observed on trees without apparent blight and ink disease symptoms. Black necroses, similar to those caused by Phytophthora infections, were noted on some of the chestnut coppices and saplings in one nursery in Ordu and led to an investigation into this disease complex. Only symptomatic plants showing dieback symptoms wer… Show more

“…DNA extractions from the isolates were performed using small amounts of mycelia scraped from the actively growing GCA cultures. PCR and sequencing of the ITS region were done using ITS 4 and ITS 6 primers, as described previously (Akıllı et al, ; Akıllı, Ulubaş‐Serçe, Katırcıoğlu, & Maden, ). Sequence analysis was carried out by the laboratory Refgen (Gen Araştırmaları ve Biyoteknoloji Ltd.…”

“…In Turkey, P. cinnamomi has been isolated from the rhizosphere soil of oaks ( Quercus petraea ) in Belgrade forest, Istanbul (Balci and Halmschlager ), and from the soil samples around the roots of symptomatic chestnut trees in five provinces of Black sea region of Turkey (Akıllı et al. ).…”

“…Molecular identification of the isolates was performed by comparison of DNA sequences produced by ITS4 and ITS6 primers as described previously (Akıllı et al. ). Isolates were grown in V8 juice (Tamek, Turkey) at 21°C for 4 days before extraction of DNA sequence analysis was carried out by REFGEN (Gen Araştırmaları ve Biyoteknoloji Ltd.…”
