Involvement of mTORC1 and mTORC2 in regulation of glioblastoma multiforme growth and motility

Abstract: Abstract. The AKT/mammalian target of Rapamycin (mTOR) signaling pathway plays a critical role in glioblastoma multiforme (GBM) oncogenesis due to activation of AKT. We studied two distinct complexes, mTOR complex 1 (mTORC1) and mTOR complex 2 (mTORC2), through which mTOR controls cell survival, growth and motility. Inhibition of mTOR by Rapamycin (RAPA) resulted in time-dependent suppression of S6 ribosomal protein (pS6K Ser235/236 ; mTORC1 substrate) and caused transient suppression of pAKT Ser473 (mTORC2 su… Show more

“…The use of inhibitors of STAT3 upstream regulators pointed that STAT3 might be involved in glioma cell migration and invasion [173,174,177,178]. This inhibition of STAT3 has been linked to down-regulation of matrix metalloproteinase gene expression such as MMP2 [179], MMP9 and to epithelial-mesenchymal transition regulating genes like Snail [180] (Figure 3).…”

“…In addition, some environmental carcinogenic factors may also be associated with the occurrence of glioma. Research has shown that electromagnetic radiation, such as mobile phone use, may be related to glioma [8]. In addition, most glioblastoma patients also have cytomegalovirus infection, and most glioblastoma specimens were found to contain virally infected macrophages [9].…”

“…In PI3K signaling cascade, mTOR acts like downstream effector molecule as well as an upstream modulator [ 40 ]. It resides in mTORC1 and mTORC2 [ 41 ]. The mTORC1 consists of mTOR, mLST8, PRAS40 and Raptor.…”
