Irradiation enhances susceptibility of tumor cells to the antitumor effects of TNF-α activated adipose derived mesenchymal stem cells in breast cancer model

Abstract: Gene modified or cytokine activated mesenchymal stem cells (MSCs) have been used as a treatment in various types of cancer. Moreover, irradiation is usually applied as either a standard primary or adjuvant therapy. Here, we showed that the expression of TNF related apoptosis-inducing ligand (TRAIL) and Dickouf-3 (Dkk-3), the promising anticancer proteins, increased in murine adipose-derived mesenchymal stromal cells (AD-MSCs) following activation with TNF-α, resulting in the induction of apoptosis in cancer ce… Show more

“…MSCs are found mainly in the connective tissue or interstitium and have marked self-renewal potential and the ability to differentiate into adipocytes, osteoblasts, and chondrocytes [6, 7]. MSCs have low immunogenicity, immunomodulatory activity, and tropism for sites of injury and inflammation and have a potential for the treatment of various diseases, including cancer [8]. These advantages have encouraged investigations of MSCs as vehicles for the delivery of anti-cancer agents [9].…”

“…Here, our current study provides novel findings, emphasizing the need for both TNBC:MSC cross-talk and pro-inflammatory signals delivered by TNFα and IL-1β, in order to achieve the most substantial levels of pro-metastatic activities: high levels of pro-metastatic chemokines, CXCL8, CCL2 and CCL5, angiogenesis, and tumor cell migration and invasion. Moreover, our findings suggest that previous studies on TNFα-treated MSCs that induced anti-tumor activities in TNBC tumors (101104) may have overlooked the actual setting that takes place in vivo , when TNBC cells interact with MSCs in the presence of TNFα stimulation.…”

“…These factors, whose production may upregulate in obesity, may be accomplices in tumor initiation. Similar to interleukin (IL)-6, IL-8, IL-1β, tumor necrosis factor-α (TNF-α), VEGF, chemokine (C-C motif) ligand 2 (CCL2) and CCL5 178, 179. Third, in obese patients, adipose tissue accommodates a higher proportion of preadipocytes, in which figures of macrophages and monocytes synergistic increase 180.…”
