IRS1 gene polymorphisms Gly972Arg and Ala513Pro are not associated with insulin resistance and type 2 diabetes risk in non-obese Turkish population

Abstract: Insulin receptor substrate 1 (IRS1), plays a critical role in insulin signaling and its control has an important place in the development of insulin resistance. The tyrosine phosphorylation of IRS1 serves as docking molecules for downstream effectors such as Phosphatidylinositol 3-kinase and phosphotyrosine phosphatase-2. We focused on the Gly972Arg and Ala513Pro variants of the IRS1 gene, since these specific allelic variants are located near the Tyr-Met-X-Met (YMXM) motifs around Tyr987 and Tyr612. Thus, we … Show more

“…However G allele carriers of IRS-1 rs2943641 have a relative risk of 4.336 (95% CI 1.592 -6.43, P = 0.006) for diabetes. Consistent with our results, in Turkish individuals there was no existed relation between this gene and T2D (37). Interestingly, in IRS-1 rs2943641 genotypes, it was observed that the individuals with BMI ≥ 25 (Kg/m 2 ) and aged > 50 years are at greater risk for developing diabetes.…”

“…So the dysregulation of IRS1 gene has an important place in the development of insulin resistance. The tyrosine phosphorylation of IRS1 serves as docking molecules for downstream effectors such as phosphatidylinositol 3-kinase and phosphotyrosine phosphatase-2 [15]. However, our study has reached no association between the Gly972Arg polymorphism and the risk of prediabetes in Hanam women.…”

“…In some studies, such a relationship was found, whereas in others, it was not. There was not significant relationship between the risk of developing type 2 DM and the IRS-1 Gly972Arg gene polymorphism in the Turkish population [59].…”
