Isolation and Partial Characterization of Bioactive Fucoxanthin fromHimanthalia elongataBrown Seaweed: A TLC-Based Approach

Abstract: Seaweeds are important sources of carotenoids, and numerous studies have shown the beneficial effects of these pigments on human health. In the present study, Himanthalia elongata brown seaweed was extracted with a mixture of low polarity solvents, and the crude extract was separated using analytical thin-layer chromatography (TLC). The separated compounds were tested for their potential antioxidant capacity and antimicrobial activity against Listeria monocytogenes bacteria using TLC bioautography approach. Fo… Show more

“…Purified fucoxanthin was determined by Fourier transform infrared spectroscopy (FT-IR). The characteristic wave number of specific functional group identified in purified sample was: OH group (3400 cm −1 ), C-H stretch (3005-2850 cm −1 ), allene (1924 cm −1 ), C═O acetate (1732 cm −1 ), conjugated C═O (1645 cm −1 ), CH 2 stretch (1605-1455 cm −1 ), geminal methyl (1380-1360 cm − 1 ), C-O acetate (1338, 1258, and 1249 cm −1 ), and trans-distributed -C═C-(1210-963 cm −1 ) which agreed with the previous reported data (Haugan et al 1992;Rajauria and Abu-Ghannam 2013).…”

“…This method was a quick-approach (Rajauria and Abu-Ghannam 2013), fast, cheap and better bioassay-directed fractionation of bioactive compounds (Hamburger and Cordell 1987) and enabled rapid progress for quick detection of new antimicrobial compounds (Suleimana et al 2010). The growth inhibition of bacteria tested was performed by the presence of clear zone around the extract against the purple background on TLC plate.…”

“…In Brazil, Peres, et al (2013) reported that ten seaweed extracts significantly inhibited the Colletotrichum lagenarium growth, but not inhibited significantly the Aspergillus flavus growth [13]. In Ireland, Rajauria and Abu-Ghannam (2013) screened antimicrobial activity and bioactive compounds of Himanthalia elongate (Brown Seaweed) [14]. In India, Indira, et al (2013) displayed the antifungal property of seaweed Halimeda tuna against nine fungal strains (Aspergillus niger, Aspergillus flavus, Alternaria alternaria, Candida albicans, Epidermophyton floccossum,Trichophyton mentagrophytes, Trichophyton rubrum, Pencillium sp.…”
