Isolation and Screening of Indigenous Fungus Producing Lignin Peroxidase from the Cocoa Plantation in Sepawon Kediri Regency Indonesia

Abstract: The aim of this research is to obtain indigenous fungus producing lignin peroxidase from the Cocoa Plantation in Sepawon Kediri Regency, Indonesia. Isolation was done using Minimum Salt Medium alkali lignin (MSM-L), purification was performed using monospore technique, lignin peroxidase activity was screened by agar medium containing methylene blue, and microscopic morphology of fungi was studied using slide culture technique. Five isolates of fungus were obtained and three isolates of fungus with different mo… Show more

“…After incubation, the appearance of a purple colour zone around the colony indicated laccase enzyme production. For lignin peroxidase activity, the method was modified from Delila et al 33 : In triplicate, the fungal plug was transferred to a PDA plate, supplemented with methylene blue (0.0065%, Sigma-Aldrich), and incubated at room temperature, in the dark, for 14 days. A clear zone around the colony, caused by the disappearance of the blue colour of the dye, confirmed the presence of peroxidase or decolorization of methylene blue dyes.…”

“…Indigenous WRF KLUM 2 isolated from the weathered cocoa husk of Sepawon plantation, Kediri, East Java has the potential as a source of ligninase since it can reduce methylene blue in selective media (Delila, 2016). Arinta (2017) showed that WRF KLUM 2 in Kirk's medium with a glucose carbon source (MK-Glucose) produced a LiP ligninase profile (60.67 U/mg) much higher than MnP (46.56 U/mg) and laccase (23.38 U/mg).…”
