Isolation, identification and characterization of phenolic acid‐degrading bacteria from soil

Abstract: Aims To isolate, identify and characterize phenolic acid‐degrading bacteria and reduce plant growth inhibition caused by phenolic acids. Methods and Results A total of 11 bacterial isolates with high phthalic acid (PA)‐degrading ability were obtained using mineral salt medium (MSM) medium containing PA as sole carbon source. These isolates were identified as Arthrobacter globiformis, Pseudomonas putida and Pseudomonas hunanensis by sequence analyses of the 16S rRNA gene. Among them, five Pseudomonas strains co… Show more

“…The DNA samples were sequenced by Genscript Biotechnology Company (Nanjing, China). The 16S rRNA gene of the isolate was amplified by PCR using universal bacterial primers F27 (5 AGAGTTTGATCCTGGCTGGCTCAG-3 ) and R1492 (5 -TACGGCTACCTTGTTACGACTT-3 ) (Wang et al, 2020). PCR amplification was performed according to the following protocol: hot start at 94 • C for 5 min, followed by 30 cycles of denaturation at 94 • C for 1 min, annealing at 55 • C for 1 min, and extension at 72 • C for 2 min, with a final extension of 72 • C for 10 min.…”

“…To identify bacteria, DNA of pre-cultured isolates was extracted using a bacterial DNA extraction kit (Magen, Guangzhou, China). Ampli cation of the 16S rRNA gene was performed using the universal bacterial primers 27f and 1492r, as described previously (Wang et al 2021). The PCR products were sequenced by the sequencing company Liuhe Huada (Beijing, China).…”
