Isolation of glioma cancer stem cells in relation to histological grades in glioma specimens

Abstract: This trend of stage-by-stage increase of gCSCs in gliomas showed statistical significance by chi-square test linear-by-linear association. We prove that the rates of existence of gCSCs increase proportionally as the WHO grades of gliomas rise.

“…This is consistent with data in the literature which indicate that the ability of CSC to differentiate into oligodendrocytes is almost null [9] or very limited [12], [23], even when markers other than GalC have been considered [16]. On the contrary, both D2 and F11 cells expressed GFAP and βIII-Tubulin at high levels.…”

“…The histological heterogeneity of tumor is consistent with reports in the literature [18], [40]. It is interesting to note that tumors have arisen in the time range reported by other Authors [7], [12], [18] and that the concentration of NS cells required to induce them is 50-fold less than that required for LI cells. Moreover, tumors in xenografts have maintained the characteristics of the original cells about expression of Nestin, GFAP and βIII-Tubulin.…”

“…After the holes were drilled on the mouse skull using a 26-gauge needle, GSC11-GFP cells (1 × 10 6 ) were implanted directly into right frontal lobe of nude mice using Hamilton syringe (Dongwoo Science Co., Seoul, Republic of Korea) at the depth of 4.5 mm. Cells were simultaneously injected into five mice using a multiple microinfusion syringe pump (Harvard Apparatus, Holliston, MA, USA) at a rate of 0.5 μl/min as previously described4748.…”

“…Herewith the content of CSCs in glioma tissue increases in proportion to the increase of the malignancy degree with a parallel increase in the expression levels of immunocytochemical markers of their stem properties [4,[7][8][9]. It is generally recognized that the identification of CSCs in various tumor types, as well as in brain gliomas is associated with a poor prognosis of the disease [10][11][12][13][14].…”
