Isolation, Synthesis, and Semisynthesis of Amaryllidaceae Constituents from Narcissus and Galanthus sp.: De Novo Total Synthesis of 2-epi-Narciclasine

Abstract: An efficient protocol for the isolation of narciclasine from common Amaryllidaceae bulbs, separation from haemanthamine, and the occurrence of a trace alkaloid, 2- epi-narciclasine, are reported. Attempts to convert natural narciclasine to its C-2 epimer by Mitsunobu inversion or oxidation/reduction sequences were compromised by rearrangement and aromatization processes, through which a synthesis of the alkaloid narciprimine was achieved. The methylation of the 7-hydroxy group of natural narciclasine followed … Show more

“…Next, the gene expression profiles of H9c2 cells after aconitine treatment were determined by NGS of mRNA on an Illumina HiSeq 4000 platform . There were 817 differentially expressed genes identified between the control and aconitine‐treated groups, including 503 upregulated and 314 downregulated genes (Figure ).…”

“…Narciprimine is 4,7-dihydroxy- [1,3]dioxolo-[4,5j]phenanthridin-6(5H)-one with a molecular weight of 271.0493 g/mol, in agreement with the molecular formula C14H9NO5 (Figure 1). It is commonly isolated in the form of white powder and crystallized from analytically pure acetic acid or acetone as fine yellowish-white needles that melt at 310-315 °C and show strong yellow-green fluorescence [7,10,15]. Although the structure of narciprimine is related to several Amaryllidaceae alkaloids, this compound displays no basic properties as its nitrogen is amidic in nature.…”
