Isoprostanes-Biomarkers of Lipid Peroxidation: Their Utility in Evaluating Oxidative Stress and Analysis

Abstract: Isoprostanes (IsoPs) are key biomarkers for investigating the role of free radical generation in the pathogenesis of human disorders. To solve IsoPs-related problems with regard to isoprostanes, analytical tools are required. This paper reviews the problems and trends in this field focusing on the methodology for assaying biomarkers in exhaled breath condensate (EBC) samples. A large amount of work has been done in the qualitative and quantitative analysis of IsoPs, but a standardized method has yet to emerge.… Show more

“…8-isoPGF 211 is a key biomarker for evaluation of free radical generation in the pathogenesis ofhuman diseases (28), and an increased lung tissue level of 8-isoPGF 211 has been reported in various pulmonary diseases (29). Production of 8-isoPGF 211 has been detected in whole lung Iysates, while inhibition of the ROCK pathway reduces the 8-isoPGF 211level in whole lung Iysates from a guinea pig model of allergy based on ovalbumin inhalation (30).…”

“…Therefore, F2-isoprostanes have been regarded as reliable biomarkers of oxidative stress. Furthermore, F2-isoprostanes have been shown to exert potent vasoconstrictor effects on animal and human vessels, suggesting a pathogenic role in cardiovascular diseases and have been extensively used as markers of lipid peroxidation in human diseases [74,75,148]. Their high stability and presence in measurable concentrations in many biological tissues and fluids, under physiological and pathological conditions, has also allowed the establishment of reference intervals and the comparison or monitoring of disease states [97,149,150].…”

“…v-3 PUFAs mainly generate F 3 and F 4 -isoprostanes [28]. F 2 -isoprostanes are considered as reliable biomarkers of lipid peroxidation in various inflammatory states, such as atherosclerosis, rheumatic diseases [48], chronic lung diseases [49], degenerative diseases, aging, obesity and cancers [50]. In obesity, one of the risk factors of CRC, F 2 -isoprostanes level is highly correlated with visceral and subcutaneous adipose tissue.…”
