Jueming prescription and its ingredients, semen cassiae and Rhizoma Curcumae Longae, stimulate lipolysis and enhance the phosphorylation of hormone-sensitive lipase in cultured rat white adipose tissue

Abstract: The present study aimed to investigate the effect of jueming prescription (JMP) and its ingredients, semen cassiae (SC) and Rhizoma Curcumae Longae (RCL), on lipolysis, and to examine their effect on the phosphorylation of hormone‑sensitive lipase (HSL) in cultured rat white adipose tissue (WAT). Retroperitoneal WAT was aseptically excised from adult male Sprague‑Dawley rats, minced into uniform sections and subjected to ex vivo culture for 24 h. The tissue sections were then distributed into a 24‑well culture… Show more

“…The adipocytes in white adipose tissue are a site of fat storage, mediated by TAG synthesis (lipogenesis) and degradation (lipolysis). Lipolysis is a biochemical process involving the breakdown of triglycerides and the release of non-esterified fatty acids and glycerol [ 135 , 136 , 137 , 138 , 139 ]. Lipolysis is catalyzed by three major enzymes: hormone-sensitive lipase, adipose triglyceride lipase, and monoacylglycerol lipase [ 135 , 137 ].…”

“…DHT stimulation of the BAT explants. BAT depots were isolated from the interscapular BAT of 6-wk-old female mice and minced for use as 1-2 mm 3 explants with minor modifications (45). The BAT explants were washed three times with PBS and placed in serum-free high-glucose DMEM for 2 h before being used for experiments.…”

“…In Utero Nicotine-Exposed Female Offspring Exhibit Increased Expression of ATGL at 6 Months, But Not at 3 Weeks Since enhanced lipolysis decreases adipocyte size (Zhang et al, 2017), we then examined whether MNE WAT had elevated expression of adipose triglyceride lipase (ATGL), the predominate enzyme involved in intracellular degradation (Zechner et al, 2009). At 6 months of age, we found a significant increase in both ATGL mRNA expression and protein levels in nicotineexposed female WAT compared with vehicle controls (Figs.…”
