Kaliziri extract upregulates tyrosinase, TRP-1, TRP-2 and MITF expression in murine B16 melanoma cells

Abstract: BackgroundKaliziri extract (KZE) is a traditional Uyghur medicine (TUM), used by traditional hospitals in China as an injection for treatment of vitiligo for more than 30 years. Clinical application has shown that this medicine has obvious therapeutic effects. However, its phytochemical analysis and mechanism have not been examined.MethodsKZE was extracted from seeds of Kaliziri [Vernonia anthelmintica (L.) Willd.] in ethanol-water (80:20, v/v), its components were identified by LC-MS/MS, and the signaling pat… Show more

“…Kaliziri is a plant that grows only in the south of Xinjiang in China, and the highlands of Pakistan and India. The extract of this plant has been found to increase tyrosinase, MITF, TRP‐1, and TRP‐2 expression [Tuerxuntayi et al, ]. Furthermore, Matsuyama et al [] studied Capparis spinose plants that were used for spices, diuretics, and hypertension treatment in ancient times and found that the plant extract could increase the melanin content and tyrosinase mRNA of B16 melanoma cells.…”

“…Butin is one of the main flavonoids in the seeds of V. anthelmintica, which for centuries has been used in Uyghur medicine for the treatment of vitiligo. 'Kaliziri' extract, from seeds of loss of V. anthelmintica, increased the expression of TYR, TRP-1, and TRP-2 via Microphthalmia transcription factor (MITF) in B16 cells (Tuerxuntayi et al, 2014); thus, we have investigated the effects of butin in the vitiligo mice model and its possible mechanisms of action.…”

“…e HEMs were treated with NaHS at the indicated concentration for 24 h and then incubated in normal medium for another 24 mentation diseases such as albinism and vitiligo. Several previous studies have shown some substances have a regulatory effect on melanin synthesis [21][22][23]. Compared with these agents, the therapeutic advantage of H 2 S is that it is a kind of small molecular weight, water-and lipid-soluble intrinsic gasotransmitter that can freely penetrate into the cell membrane to exert a biological effect.…”
