“…As Kaposi's sarcoma is infrequent in Pacific Islands, classical immunohistochemistry was performed on deparaffinised sections, using a panel of antibodies including anti-factor VIII, anti-CD31 and anti-CD34 antibodies as well as proliferation cell nuclear antigen antibodies (PCNA) and Ki67, as previously described (Hbid et al, 2005). The HHV-8-specific antibody directed against latent nuclear antigen (LNA-1 encoded by open reading frame 73), was provided by Novocastra (clone 13 B10) and used at a dilution of 1:30, as described elsewhere (Hbid et al, 2005). Two different immuno-fluorescence assays (IFA) were used, as previously described (Kedes et al, 1996;Plancoulaine et al, 2002): (i) a commercial test (HHV-8 immunofluorescence antibody assay; ABI, Columbia, MD), using the KS-1 cell line as a HHV-8 source of antigens and detecting antibodies mainly directed against lytic HHV-8 antigens and (ii) an in house test using BC-3 cells which express only HHV-8 latent antigens (LNA).…”
