Karyotype and C-banding patterns of mitotic chromosomes in Henrardia persica (Boiss.) C.E. Hubb

Abstract: Abstract·The C-banded karyotype of Henrardia persica (2n=2x=14), an annual self-pollinated grass species was studied in two natural populations using aceto-iron-hematoxilin staining and C-banding technique. It was revealed that the karyotype of this species is asymmetric and consists of 4 pairs of sub-acrocentric and 3 pairs of acrocentric chromosomes. One of chromosomes has a satellite located on the end of its long arm. Each chromosome having a unique C-banding pattern is easily recognizable.

“…The root tips were pretreated in 0.05% solution of colchicine for 2.5 h at room temperature before they were fixed in chromic acid -formalin (1:1 v:v) at 4°C for 25 h. Staining with hematoxylin was done as described earlier by Asghari-Zakaria et al (2002).…”

“…The root tips were hydrolyzed by NaOH 1 N for 8 min in a water bath at 60 C and then stained with ferric hematoxylin solution for 16 h at room temperature. To complete softening of the root tissue, the root tips were treated in cytase enzyme for 2 h (Asghari-Zakaria et al 2002). Metaphase cells were observed with a Leica (Leica Co.) microscope model Gallen III.…”
