Karyotyping of Candida albicans and Candida glabrata isolates from recurrent vaginal infections by pulsed-field gel electrophoresis

Abstract: In the present study, 16 women with recurrent vulvovaginal candidiasis (RVVC) due to Candida albicans and Candida (Torulopsis) glabrata were followed for a period of 4 to 12 months, and 36 vaginal isolates were evaluated by pulsed-field gel electrophoresis (PFGE). Eleven women were infected by C. albicans and 5 by C. glabrata. Three electrophoretic karyotypes of C. albicans and 3 of C. glabrata were identified throughout the follow-up. All patients but one was infected with the same karyotype of C. albicans or… Show more

“…On the other hand, samples taken from the same NN at different times during the hospitalisation showed the same pulsotype. Our findings are in agreement with prior studies in patients with invasive or tegumentary candidosis; and it is accepted that, in most cases, the invasive and colonising strains are identical 12,23,24,28,31 …”

“…13,15,21 This polymorphism is reflected by the large variability of the pulsotypes reported in previous epidemiological studies carried out both in hospitalised patients and in patients with superficial candidosis. 17,20,[22][23][24][25][26] Systemic candidosis in NN has two possible sources, either endogenous or exogenous. 1,19,[27][28][29] In the last situation, nurses represent an important relay in the contamination of NN.…”

“…The average fre- quency of candidaemia episodes in our university, 0.41 per 10,000 patient days, was lower than the corresponding data reported from Spain (0.73), Italy (0.90) and Ireland (0.70), but similar to that observed in a European survey (0.44) [4,5,21,22]. The most frequent causative agent each year was C. albicans, as reported similarly from other countries both inside and outside Europe [6,11,19,20,[22][23][24]. “…Variations of karyotypes between C. glabrata strains have been previously observed, but concern mostly the two largest chromosomes, XII and XIII (Barchiesi et al, 1999;Doi et al, 1994;Fodor et al, 2002;Kaufmann and Merz, 1989;Khattak et al, 1992;Magee and Magee, 1987;Schwab et al, 1997;Vazquez et al, 1993), that bear the repeated rDNA loci known to be variable in copy number (Asakura et al, 1991;Dujon et al, 2004). We have re-examined this question in a more extensive manner using a collection of 183 C. glabrata strains, isolated mainly from patients, with three strains from the environment (see Section 2).…”
