Kazachstania siamensis sp. nov., an ascomycetous yeast species from forest soil in Thailand

Abstract: Two strains (S-34T and S-35) of a novel ascomycetous yeast species belonging to the genus Kazachstania were isolated from soil from a mixed deciduous forest in Amphoe Wang Nam Khiao, Nakhon Ratchasima Province, Thailand. The D1/D2 domains of the large-subunit rDNA sequences of the two strains were identical and also indicated a close relationship with respect to Kazachstania aquatica, Kazachstania unispora, Kazachstania aerobia, Kazachstania servazzii and Kazachstania solicola. The most closely related species… Show more

“…The methods for DNA extraction and amplification of the D1/D2 region of the LSU rRNA gene and the ITS region were as described previously (Limtong et al, 2007). The PCR product was checked by agarose gel electrophoresis and purified using a HiYield Gel/PCR DNA Fragments Extraction kit (RBC Bioscience), according to the manufacturer's protocol.…”

“…Ubiquinones were extracted from cells cultivated in 500 ml Erlenmeyer flasks containing 100 ml YPD broth on a rotary shaker at 28 C for 48 h and purified according to the methods described by Yamada & Kondo (1973) and Kuraishi et al (1985). Isopreologues were identified by HPLC as described previously (Limtong et al, 2007). …”

“…The type strain is RV5 Yeasts in the genus Candida are distributed widely in both terrestrial and aquatic habitats. In aquatic habitats, Candida strains have been found in fresh water, estuarine water and sea water (Cooke et al, 1960;Hagler & Ahearn, 1981Spencer & Spencer, 1997;de Almeida, 2005;Butinar et al, 2005;Nagahama, 2006;de García et al, 2007;Limtong et al, 2007Limtong et al, , 2008Boonmak et al, 2009;Limtong et al, 2010). Mangrove forests, or intertidal forests, are plant communities in saline coastal habitats distributed over a wide range of geographical zones (Aksornkoae, 1999).…”

“…In total 54 grape leaf samples were collected with 8, 24 and 22 samples collected on 4 February 2009, 2 July 2009 and 2 September 2009, respectively. Yeasts were isolated from the surface of plant leaves by an enrichment technique using yeast extract malt extract (YM) broth (0.3% yeast extract, 0.3% malt extract, 0.5% peptone and 1% glucose) supplemented with 0.025% sodium propionate and 0.02% chloramphenicol (Limtong et al, 2007). Three grams of cut leaves were aseptically placed in 250 ml Erlenmeyer flasks containing 50 ml enrichment broth and incubated on a rotary shaker at room temperature (27 3 C) for 2 days.…”

“…The sequences of the D1/D2 domain of the LSU rRNA gene and the ITS region were determined from PCR products amplified from genomic DNA. Methods for DNA extraction and amplification of the D1/D2 domain of the LSU rRNA gene have been described previously (Limtong et al, 2007). The ITS region was amplified with primers ITS1 and ITS4, following the method of White et al (1990).…”
