Kidney injury and cell therapy: Preclinical study

Abstract: The aim of this study is to show histological and immunofluorescence analysis of renal parenchyma of agoutis affected by gentamicin-induced renal disease after the infusion of bone marrow mononuclear cells (BMMC) stained with Hoechst®. Nine agouti's males were divided into three groups: Test group (TG): renal disease by gentamicin induced (n = 3), cell therapy group (CTG): renal disease by gentamicin induced and BMMC infusion (n = 3), and control group (CG): nonrenal disease and BMMC infusion (n = 3). TG and C… Show more

“…The agouti is a rodent that has been extensively studied , Menezes et al 2003, Azevêdo et al 2008) and which shows biological and physiological characteristics favorable for use in experimental research as a animal model (Fortes et al 2005, Conde-Júnior et al 2012, Rocha et al 2012. Bone marrow mononuclear cells of this animal were injected to treat induced renal injury (Cabral et al 2012) and in culture undifferentiated cells were obtained through the isolation and expansion by several passages (Rocha et al 2012). Despite these preliminary studies involving agouti stem cells, there are few reports in the literature, and therefore, in order to verify the potential of mesenchymal stem cells from other niches of this animal, the objective of the present study was to characterize the behavior of stem cells obtained from incisor tooth pulp in culture.…”

“…A study in rats with mesenchymal stem cell therapy for acute kidney injury induced by ischemia/reperfusion model therapy accelerated renal tissue recovery 7 . The same occurred in a wild rodent (agouti -Dasyprocta prymnolopha) model, in chronic renal injury induced by drugs 8 .…”

“…This animal is being a rich source of protein and hunting target in the region, but is not on the list of endangered animals (BRASIL, 2016). In this way, its production in captivity is expanding, since several research groups are developing studies on its morphology, ultrasonographic and echocardiographic evaluation, as well as a biological model for alternative treatments of chronic diseases (CABRAL et al, 2012 ;CARVALHO et al, 2015 ;DINIZ et al, 2013 ;FORTES et al, 2013;SILVA et al 2014 ;SOUSA et al, 2013). Blood is responsible for transporting and micronutrient elements related to metabolism and thus its analysis reflects the physiological situation of individual.…”
