Kinetics of Nuclear Status and Kinase Activities During In Vitro Maturation of Canine Oocytes

Abstract: Abstract. In contrast to those of other mammals, canine oocytes are ovulated at the germinal vesicle (GV) stage and then progress to the metaphase II (MII) stage in the oviduct. In other species, oocytes at the MII are widely used for in vitro fertilization or as recipients in somatic cell nuclear transfer. Many researchers have tried to improve the in vitro maturation (IVM) of canine oocytes. However, the proportion of MII oocytes remains low, resulting in poor efficiency of embryogenesis in vitro. This leads… Show more

“…Moreover, our data underpin that the optimal culture period for IVM of bitch oocytes is 72 h, and are in accordance with those previous reported (Otoi et al. 2004; Suzukamo et al. 2009).…”

“…Overall, 2,980 oocytes (1,470 from diestrus phase and 1,510 from anestrus) with the zona pellucida were transferred to culture plates with 0.2% hyaluronidase solution for removal of cumulus cells. Subsequently, they were washed several times with PBS and placed into cell lysis buffer composed of 20 mM TRIS, 150 mM NaCl, 1 mM EDTA, 1 mM EGTA, 1% Triton X‐100, 2,5 mM Sodium phosphate, 1 mM β‐glycerophosphate, 1 mM Na 3 VO 4 , 1 mg/ml leupeptin and 1 mM phenylmethylsulfonyl fluoride (Suzukamo et al, ). The lysis solution was added to the sample at the ratio of 1:1, that is, 1 oocyte/ 1 μl of solution.…”

“…Enzyme digestion was performed overnight in humid chamber at 37°C, and thereafter, the enzyme was inactivated by adding 1% trifluoroacetic acid to the mixture (final concentration). Desalination was performed by the STAGE‐TIP method using C18 membrane (3M Empore; Suzukamo et al, ).…”

“…In connection with differences in specific of oocyte maturation there are many problems with effective culture of these gametes (Suzukamo et al 2009). Until now there have been many studies performed on maturation and related with rearrangements within the oocyte and cumulus cells using in vitro model.…”

“…Until now there have been many studies performed on maturation and related with rearrangements within the oocyte and cumulus cells using in vitro model. Suzukamo et al (2009), using mitogen, activated kinase (MAPK) and p34 kinase, typical markers of development demonstrated that the optimal oocyte culture time is 72 h, because at this time MAPK and p34 reveled the highest activity. In contrast to these results, culture performed with the combination of chemical and biological agents allowed to obtain oocytes in meiosis II after 48 h (Hanna et al 2008).…”
