Kinetin Is the Most Effective Cytokinin on Shoot Multiplication from Cucumber

Abstract: In vitro shoot multiplication of cucumber (Cucumis sativus) was examined from the nodal explants of 10-day-old aseptic plantlets using Murashige and Skoog (MS) media supplemented with different concentration (0, 0.5, 1, 2, 3 mg L -1 ) of cytokinins (6-Benzylaminopurine-BAP, Kinetin-Kn, Thidiazuron-TDZ, and Zeatin). Nodal explants of cucumber showed shoot induction and multiplication in response to all cytokinins tried. MS medium containing Kn was the most effective for inducing shoots from nodal explants of cu… Show more

“…For example, the multiplication of a medicinal plant (Scopari dulcis) showed that shoot induction was improved by a combination of both BAP and KIN (Premkumar et al, 2011). On the contrary, for cucumbers, the best results of the shoot multiplication stage were obtained when only kinetin was supplemented to the MS medium (Abu-Romman et al, 2015).…”

“…Cytokinins are commonly used in plant tissue culture to initiate cell growth and also to induce the formation of shoots whereby the are actively take part in various plant physiological processes ( Davies 1995 ). This includes shoot multiplication ( Kaviani et al 2013 ; Bhat et al 2013 ; Abu-Romman et al 2015 ), tuberous root production ( Deshwal & Trivedi 2011 ), callus induction ( Borjian & Arak 2013 ), leaf senescence ( Riefler et al 2006 ) and shoot apical meristem activity regulators ( Tucker & Laux 2007 ). Cytokinin such as kinetin is known to initiate cell division in the presence of auxin and is widely used together with auxins for callus formation or to induce shoots where low levels of auxin is available.…”

“…However, HgCl 2 solution is highly toxic and it is therefore recommended for frequent use in sterilization procedure. Growth regulators especially cytokinins are one of the most important factors affecting the regeneration of microshoots [30][31][32][33]. Kinetin, BAP, TDZ and Zeatin have been used in cassava micropropagation [17,[34][35][36].…”
