KLK3, PCA3, and TMPRSS2-ERG expression in the peripheral blood mononuclear cell fraction from castration-resistant prostate cancer patients and response to docetaxel treatment

Abstract: The feasibility of a highly sensitive modified nucleic acid amplification assay to assess KLK3, PCA3, and TMPRSS2-ERG mRNA in the PBMC fraction from CRPC patients was demonstrated. Moreover, response of these markers to systemic treatment was shown.

“…The scant data published so far suggest that a correlation exists between messenger ribonucleic acid (mRNA) expression of AR (AR splice variant 7 messenger RNA, AR-V7) in circulating tumor cells and PCa progression after enzalutamide or abiraterone treatment and survival in patients with PCa [9]. A recent study confirmed the expression of KLK3 (kallikrein-related peptidase 3) in peripheral blood samples from patients with clinically localized PCa (T1-T2) [10].…”

“…Regarding other genes, KLK3, PCA3, and TMPRSS2-ERG mRNA expression levels are increased in most castration-resistant patients with PCa (CRPC); accordingly, these genes have been proposed as biomarkers for the prognosis of castration-resistant patients with PCa or to measure therapy effects or both [10]. mRNA expression levels in the TMPRSS2-ERG fusion gene and ERG have been found to be associated with an aggressive phenotype and have a prognostic value and proved to serve as molecular markers of PCa progression [15].…”

“…Both unfavorable CTC count (five or more cells) and detection of two or more gene transcripts had similar significant prognostic value for risk of PCa death, and when combined, additional prognostic value was demonstrated. With a similar approach, kallikrein-related peptidase 3 (KLK3), PCA3, and TMPRSS2:ERG mRNA could be detected in the peripheral blood of CRPC patients but not in healthy controls [93]. Also, decreased expression levels of these genes were noted after docetaxel treatment, suggesting a potential role for treatment monitoring.…”

Additionally, KLK3, PCA3 and TMPRSS2:ERG mRNA could be detected in the peripheral blood of CRPC patients but not in healthy controls [143]. Decreased expression levels of these genes, however, were noted after docetaxel treatment, suggesting a potential role for treatment monitoring.…”
