Knockdown of STAT3 targets a subpopulation of invasive melanoma stem‐like cells

Abstract: Transcription factor signal transducer and activator of transcription 3 (STAT3) is constitutively activated in many cancers, including melanomas. Active, phosphorylated STAT3 contributes to tumor growth and formation of the immunosuppressive tumor microenvironment. Recent evidence suggests an important role of STAT3 in self‐renewal of cancer stem‐like cells (CSCs). In the present study, we aimed to determine the expression and role of active STAT3 in melanoma CSCs. We found the increased levels of phosphorylat… Show more

“…Mechanistically, the present study shows that NP attenuates MCF-7-R cell stemness through suppressing STAT3 transcriptional activity. STAT3 transcription activity regulates the proliferation, survival, apoptosis and drug resistance of cancer cells and CSCs [21,22]. Previous studies have shown that STAT3 is indispensable to these properties and functions of CSCs, and activation of STAT3 (phosphorylation of Y705 and S727) triggers gene expression that controls CSCs progression.…”

“…STIP1 stimulates the expression of Janus kinase 2 (JAK2), which activates STAT3 ( Figure 1 ) [ 64 ]. STAT3 plays an important role in self-renewal of MM stem-like cells [ 65 ]. Accordingly, targeting STAT3 sensitizes human MM cells to the BRAF inhibitor vemurafenib [ 66 ].…”

“…Previous studies have demonstrated that STAT3 is overexpressed in many types of tumor cells (Misra et al, 2018;Shastri et al, 2018;Kulesza et al, 2019). To determine whether the STAT3 pathway is required for SNX-2112-induced ECSLCs apoptosis, clinical esophageal cancer samples were collected and the STAT3 and p-STAT3 expression levels were evaluated using IHC.…”

“…Accumulating evidence has demonstrated that STAT3 signaling is associated with the proliferation of CSCs or TICs (Chung et al, 2013;Kulesza et al, 2019). STAT3 is a key transcription factor with many functions in normal stem cells, CSCs, and embryonic stem cells (ESCs) (Zhong et al, 1994;Marotta et al, 2011;da Hora et al, 2019;Zhang et al, 2019).…”
