Lack of association between paraoxonase-1 Q192R polymorphism and rheumatoid arthritis in southeast Iran

Abstract: ABSTRACT. Decreased paraoxonase-1 (PON1) activity has been associated with rheumatoid arthritis. There are two polymorphisms in serum PON1; one differs in the amino acid at position 192 (Q192R) and the other one differs at position 55 (L55M). We looked for a possible association between Q192R polymorphism and rheumatoid arthritis. The Q192R polymorphism in 88 rheumatoid arthritis patients and 78 healthy subjects was determined using tetra amplification refractory mutation system-polymerase chain reaction (ARMS… Show more

“…The local Ethics Committee of the Zahedan University of Medical Sciences approved the project, and written informed consent was obtained from all participants. Genomic DNA was extracted from whole blood samples, as described previously (Hashemi et al, 2010a). SNP genotyping of P2X7R was performed using tetra-amplification refractory mutation system-polymerase chain reaction (T-ARMS-PCR), which is a simple and rapid method for detecting SNP (Hashemi et al, 2010b.…”

“…The SNPs replace a nucleotide with another nucleotide in gene structure. When SNPs arise within a gene or in a regulatory region near a gene, they may play a direct role in disease by affecting the gene's function (14,15). If a SNP take place in coding regions of genes, it causes replacement of an amino acid in a protein structure with another amino acid, which results in the alteration of protein activity (16).…”

“…The formation of free radicals is a normal outcome of a variety of essential biochemical reactions and can occur at elevated rates under pathophysiological circumstances (12,13). The physiological role of antioxidants is to prevent damage to (16,17). It has been well documented that the two common coding region polymorphisms of the gene PON1 (L55M and Q192R) lead to changes of both the level and activity of the enzyme (18,19).…”
