Landiolol decreases a dysrhythmogenic dose of epinephrine in dogs during halothane anesthesia

Abstract: Purpose: To examine the effect of landiolol (ONO-1101), a new ultra-short acting and highly selective beta~ blocker, on epinephrine-induced ventricular arrhythmias in halothane-anesthetized dogs. Methods: We administered five different doses (0, 0. I, 0.5, 1.0, 10/Jg'kg-I'min -0 landiolol and determined the dysrhythmogenic dose of epinephrine (DDE), defined as the smallest dose producing four or more PVCs within 15 sec, at each dose of landiolol and after cessation of infusion. l~.sults: The control value of D… Show more

“…We have previously demonstrated that epinephrine causes dysrhythmias in halothane-anesthetized dogs, and that 10 µg·kg -1 ·min -1 landiolol prevents such dysrhythmias. 17 In this study, the dose of 1 µg·kg -1 ·min -1 landiolol controlled dysrhythmias associated with theophylline intoxication.…”

“…In fact, most beta-blockers, including landiolol, have antiarrhythmic properties. Landiolol decreased the dysrhythmogenic dose of epinephrine in dogs [12], and restored sinus rhythm in patients with supraventricular or ventricular tachycardia [2]. Although there is no case report describing the conversion of atrial fibrillation due to landiolol, esmolol was reported to accelerate the conversion of recent-onset atrial fibrillation compared with diltiazem and verapamil [13,14].…”

“…7,8 While landiolol is more potent than esmolol 7 in terms of chronotropy, it also has less effects on blood pressure than esmolol in anesthetized rabbits. 9 Although there have been great advances in cardiac surgical techniques in the past decade, postoperative support with inotropic agents is still frequently necessary.…”

“…Animal preparation was similar to that described previously. 8,13 Animals were anesthetized with 25 mg·kg -1 pentobarbital iv, the trachea intubated with a cuffed tracheal tube, and the lungs ventilated mechanically. Anesthesia was maintained with 1. were randomly assigned according to computer-generated random numbers into one of two groups (n = 9 in each): 1) a milrinone group, or 2) a control (normal saline) group.…”
