Large‐scale production of wheat and triticale double haploids through the use of a single‐anther culture method

Abstract: A total of 257 parental wheat and 38 triticale lines were used for anther culture. On average, 2.1 green wheat haploids were obtained per spike. This response occurred irrespective of the origin of the material (Germany, France, Sweden or UK) and 5 years of testing. Triticale responded with 5.3 green haploids per spike. Using the criterion that one parental line should give at least one green haploid per spike in the screening experiment, green haploids were produced from 88 out of 91 F1 wheat breeding combina… Show more

“…Cold pretreatment is a common system to induce embryogenesis. In the present study, some of cultivars after pretreating their tillers at 4°C for 21 days, showed a proper response to microspore culture that was consistent with findings of Armstrong et al (1987) and Tuvesson et al (2000). Cold pretreatment can increase the total content of free amino acids which cause to adaptation of microspores to the metabolic changes and induction of embryogenesis (Claparols et al, 1993;Xie et al, 1997).…”

“…The efficiency (green plantlets production/100 anthers) of AC was screened in different winter wheat breeding programmes and the mean of the analysis was 0.4-5.3 green plantlets/100 anthers (Holme et al, 1999;Tuvesson et al, 2000;Lantos et al, 2013). Because of genotype dependency, this value was higher (more than 100 green plantlets/100 anthers) in AC of some high-responding genotypes (Broughton, 2011;Lantos et al, 2013;Castillo et al, 2015).…”

“…Anther culture of cv. Bogo has shown variable success: early attempts resulted in very low plant yields or only albino plants (Rogalska and Mikulski 1995;Golemiec and Wędzony 2000), while Tuvesson et al (2000) reported 4.4 green haploids per spike, Wędzony et al (2000) obtained five green plants per 100 plated anthers and Zimny (1997) reported up to 10.2 green plants per 100 anthers of Bogo transgenics. In comparison, plant yield per spike in the isolated microspore cultures described herein was essentially an order of magnitude higher than the best results of anther culture.…”
