Latanoprost (LAT) has been shown to have a hypertrichotic effect, which makes it a promising candidate for alopecia treatments. For the first time, LAT has been encapsulated in nanotransfersomes in order to increase its efficacy. Ex vivo skin biodistribution was studied by confocal laser microscopy both in human scalp and pig skin. Results showed that nanotransfersomes increase the penetration of two different fluorochromes, with similar patterns in both species, compared with fluorochrome solutions containing no nanotransfersomes. Nanotransfersomes were stable under accelerated conditions (40 °C/75% RH) and long-term conditions (25 °C/60% RH) for up to 1 year, with no differences in vesicle size and polydispersity when LAT was loaded. Nanotransfersomes increased the LAT cell proliferation effect in HaCaT cell via MAPK signaling pathway. Collectively, our results demonstrate LAT-nanotransfersomes formulation could be a promising therapy for hair growth disorders.

INTRODUÇÃO: A latanoprosta é um análogo à prostaglandinas utilizadas no tratamento de glaucoma, mas que tem sido usado para avaliação da hipertricose. OBJETIVO: Revisar através da literatura o papel da latanoprosta no crescimento de pelos. METODOLOGIA: Trata-se de uma revisão descritiva que avaliou publicações indexadas na base de dados MEDLINE, consultadas por meio do PUBMED; Biblioteca Virtual de Saúde (BVS), Periódicos Capes e Scientific Eletronic Library Online (SCIELO), nos meses de junho e julho de 2018. A partir das análises foram incluídos apenas trabalhos que fizeram uso da latanoprosta em humanos. RESULTADO E DISCUSSÃO: De 111 artigos relacionados com a latanoprosta e o crescimento de pelos, apenas nove foram incluídos. A maioria dos estudos eram do tipo caso-controle com avaliação do uso da latanoprosta principalmente no crescimento de pelos do couro cabeludo e cílios tratados para alopecia areata e androgenética. O número de participantes dos estudos variou de 1 a 54 indivíduos. A latanoprosta foi utilizada principalmente por via tópica na concentração de 0,005%. Infelizmente nem todos os trabalhos incluídos avaliaram a eficácia da latanoprosta ou descreveram adequadamente a maneira como foi investigado o crescimento dos pelos. CONCLUSÃO: Apesar de não serpossível afirmar que a latanoprosta é eficaz como terapia capilar, este estudo é promissor e pode apoiar novos trabalhos que investiguem melhor a eficácia e segurança da latanoprosta, bem como a dose ideal e duração do tratamento dela ou de outro candidato análogo das prostaglandinas.

Utilising SNP Association Analysis as a Prospective Approach for Personalising Androgenetic Alopecia Treatment

Introduction Androgenetic alopecia (AGA) is a prevalent, multifactorial form of hair loss involving complex aetiological factors, such as altered androgen regulation and energy metabolism. Existing treatments offer limited success, thus highlighting the need for advanced, personalised therapeutic strategies. This study focuses on correlating the genetic mechanisms of AGA with molecular targets involved in the response to current treatment modalities. Methods An anonymised database including 26,607 patients was subjected to analysis. The dataset included information on patients’ genotypes in 26 single nucleotide polymorphisms (SNPs), specifically, and diagnosed AGA grades, representing a broad range of ethnic backgrounds. Results In our sample, 64.6% of males and 35.4% of females were diagnosed with female pattern hair loss. This distribution aligns well with prior studies, thus validating the representativeness of our dataset. AGA grading was classified using the Hamilton–Norwood and Ludwig scales, although no association was found to the grade of the disease. SNP association analysis revealed eight SNPs, namely rs13283456 (PTGES2), rs523349 (SRD5A2), rs1800012 (COL1A1), rs4343 (ACE), rs10782665 (PTGFR), rs533116 (PTGDR2), rs12724719 (CRABP2) and rs545659 (PTGDR2), to be statistically significant with a p-value below 0.05. Conclusions The study establishes a preliminary association between eight specific SNPs and AGA. These genetic markers offer insights into the variability of therapeutic responses, thus underlining the importance of personalised treatment approaches. Our findings show the potential for more targeted research to understand these SNPs’ and further roles in AGA pathophysiology and in modulating treatment response.
