LC‐MS based metabolomics identification of novel biomarkers of tobacco smoke‐induced chronic bronchitis

Abstract: Tobacco smoke (TS) is a major causative agent to lead to chronic bronchitis (CB). However the mechanisms of CB induced by TS are unclear. In this report, rats were exposed to different concentrations of TS and the metabolic features of CB were characterized by using a nontargeted metabolic profiling method based on liquid chromatography-mass spectrometry (LC-MS) to detect the altered metabolic patterns in serum from CB rats and investigate the mechanisms of CB. 11 potential biomarkers were identified in serum … Show more

“…Metabolomics is an emerging technology that is being used to identify new biomarkers of tobacco smoke exposure (117125), and for studying COPD (126128). The assay can be used to identify thousands of small molecules (<1500 Daltons) reflective of exogenous exposures and cellular responses to those exposures.…”

“…Lysophosphatides include LPC, LPE and lysophosphatidic acid (LPA), which are converted from phospholipids through the hydrolysis by the enzyme of phospholipase A2 (PLA2) (Ren et al, ). Owing to different replacement groups, phosphoglyceride can be divided into different classes, such as phosphatidylcholine and phosphatidylethanolamine .…”

“…5‐Hydroxytryptamine (5‐HT) is a product of tryptophan through the reaction of hydroxylation and decarboxylation, which is involved in the response of inflammation and pain. 5‐HIAA, as a derivative of 5‐HT, is also involved in the inflammation process (Ren et al, ). It was considered that 5‐HT and its metabolite 5‐HIAA were associated with suicidal/aggressive behavior, which had been described as the major neurochemical system in the patient's brain with major depression (Hamon and Blier, ).…”

“…Mass spectra of these unavailable biomarkers were identified using the accurate mass and MS/MS fragments with searching in reliable biochemical databases such as the Human Metabolome Database (HMDB; http://www.hmdb.ca/) and the Kyoto Encyclopedia of Genes and Genomes (KEGG; http://www. http://kegg.com/) (Ren et al, ; Wang et al, ). The pathway analysis of potential biomarkers was performed with MetaboAnalyst based on database source including HMDB and KEGG.…”
