Grapevine (Vitis spp.) is one of the most important socio-economically important plants in the global scale, and the need for its breeding is increasing. In viticulture, mutation is more promising than crossbreeding in breeding new genotypes from natural genetic diversity. Polyploid plants outperform their diploid relatives in several respects. In this study, the efficacy of oryzalin and N2O mutagens in the induction of polyploidy was investigated by applying different doses and durations to the forced cuttings of 41B [Chasselas (Vitis vinifera L.) × Vitis berlandieri Planch)] and Fercal [(Vitis vinifera x Vitis berlandieri) × 333 EM] rootstocks. LD50 values of mutagen applications were determined, morphological and cytological effects were examined by macroscopic, microscopic, and cytological methods. Application time and dose increase of mutagens decreased LD50 values. As a result of mutagen applications, leaf thickness and chlorophyll content of the surviving plants increased. Applications increased stomatal sizes, decreased their density, increased chloroplast numbers, increased leaf thicknesses and partially SPAD values. It was determined that they were not polyploid in the confirmation test performed with flow cytometry (FC) analyses in 4 Fercal and 1 41B samples that were assumed to be mutant by stoma and chloroplast examinations. After that, it was thought that it would be appropriate to try vegetative material with actively dividing cells, such as nodal cuttings, in the studies of obtaining polyploid individuals on grapevine rootstocks

Polemonium caeruleum L. (Polemoniaceae) is a valuable medicinal herb with a wide spectrum of biological activities. Under natural conditions, the productivity of this species is rather low. In this study, colchicine-induced tetraploid plants (2n = 4x = 36) of P. caeruleum were obtained, and for the first time, their morphological and cytogenetic characterization was performed. In the tetraploid plants, raw material productivity and also the content of triterpene saponins were significantly higher than in the control diploids. The analysis of chromosome behavior at meiosis and FISH chromosome mapping of 45S and 5S rDNA generally demonstrated stability of both genomes in the tetraploid plants. Based on chromosome morphology and distribution patterns of the studied molecular cytogenetic markers, all chromosome pairs in karyotypes were identified, and chromosome karyograms and idiograms of P. caeruleum were constructed. The revealed specific microdiagnostic characteristics of P. caeruleum (strongly sinuous cells and anomocytic stomata of the leaf epidermis, and also glandular hairs along the veins) could be useful for raw material identification. In the obtained tetraploids, the predominance of large stomata on the lower leaf epidermis was determined. The studied tetraploids can be used in various breeding programs to obtain high-quality pharmaceutical raw materials of P. caeruleum.

Sodium azide mempunyai kapasitas dalam induksi perubahan genetik dalam mengindikasikan kapasitas hasil tanaman dan sensitivitas terhadap kondisi lingkungan tercekam. Penelitian ini bertujuan untuk mengetahui respon padi inpari dengan perlakuan lama perendaman pada mutagen sodium azide terhadap sensitivitas, karakter bibit, dan jarak genetik berdasarkan karakter morfologi. Percobaan disusun menggunakan rancangan petak terbagi dengan varietas padi (Inpari 31, 32, dan 33) sebagai petak utama, dan lama perendaman mutagen sodium azide (0, 5, 10, 15, dan 20 jam) sebagai anak petak. Hasil menunjukan bahwa Inpari 32 lebih sensitif (LD50), dan lebih baik pada karakter daya kecambah dan karakter bibit dibandingkan dengan Inpari 31 dan 33. Jarak genetik terdekat ditunjukan antara Inpari 31 dan Inpari 33 tanpa mutagen, dan antara Inpari 32 dan Inpari 33 dengan perendaman sodium azide selama 10 dan 20 jam.
