This study aimed to observe the viability of the pollen grains of Malpighia emarginata DC. (West Indian cherry) and the action of a lectin in their germination. Lectins are proteins that specifically interact with carbohydrates, but don't modify them and bind with high affinity and specificity, promoting a transfer of information that is clearly central to many cellular processes in living beings. For the viability test was used aniline blue in lactophenol. The in vitro germination test used was the hanging drop method, in control medium containing sucrose, boric acid, calcium nitrate and agar. The experiment dealt with three different treatments in order to pollen germination: growing medium without addition of lectin, with 1.0 µg/ml and with 3.0 µg/ml CPL lectin. This lectin is extracted from seeds of Crotalaria pallida L.-Leguminosae. Data on pollen grains were analyzed by one-way ANOVA, with 95% reliability and comparison of averages by Turkey test at 5% probability. It was found that despite previous high viability of the pollen grains (c. 70%), a germinability rate has been low, yet exceptionally larger in pollen samples placed on a growing medium with addition of 3.0 µg/ml of CPL lectin (24%). There was no significant difference among the pollen grains germinate in medium without lectin and those in medium with the addition of only 1.0 µg/ml. According to data obtained in the treatments, CPL lectin, with concentration of 3.0 µg/ml, influenced the formation of the pollen tube and thus more pollen germinated in Malpighia emarginata.

RESUMOLectinas são proteínas que se ligam com afinidade a resíduos de glicídios com capacidade de aglutinar células e preciptar glicoconjugados. Essas proteínas são importantes porque estão envolvidas no processo de reconhecimento celular e sinalização em diversas vias metabólicas. O objetivo deste estudo foi isolar, purificar e investigar a atividade biológica das lectinas de Crotalaria spectabilis R. A extração foi feita das sementes, as quais foram moidas e suas células lisadas em solução NaCl 0,15 M. Seguiu-se precipitação protéica por acetona e sulfato de amônio. A partir dos dados da quantificação, observou-se que o método de precipitação salina apresentou maior rendimento de proteínas. Os ensaios de hemaglutinação foram positivos para atividade hemaglutinante. Assim, a partir dos dados obtidos concluímos que nas sementes de C. spectabilis R. existem lectinas com capacidade de reconhecer os receptores presentes na membrana dos eritrócitos humanos ligando-se a estes e promovendo a aglutinação celular. Palavras-chave:Especificidade, Hemaglutinação, Receptores. INTRODUÇÃOLectinas são proteínas com capacidade de se ligarem especificamente a carboidratos e aglutinarem células. Nas plantas estão presentes em todos os órgãos, embora encontradas em maior quantidade nas sementes de leguminosas. Apresentam distribuição e atividade em todos os organismos vivos ocorrendo desde vírus até mamíferos 1,2 , embora tenham sido pesquisadas em maior número em plantas 3,4 . O objetivo da pesquisa foi isolar, purificar e investigar a atividade biológica das lectinas extraídas de sementes de Crotalaria spectabilis R., para que possam ser utilizadas como uma ferramenta molecular em áreas como oncologia, bacteriologia, virologia entre outras aplicações. Dessa forma, pesquisas sobre a atividade biológica de lectinas das sementes de Crotalaria spectabilis R. são importantes para a compreensão de processos envolvidos no reconhecimento celular, para que funcionem como uma ferramenta molecular em novos estudos, assim como na descoberta de produtos biotecnológicos inovadores.

RESUMODiferentes classes de proteínas são comuns em sementes de leguminosas, incluindo inibidores de tripsina e proteínas hemaglutinantes, as quais atuam sobre enzimas proteolíticas e sobre carboidratos da superfície celular, respectivamente. O objetivo deste trabalho foi quantificar, detectar e caracterizar parcialmente a ocorrência dessas proteínas em sementes de Tachigali plumbea, Sesbania exasperata e Ormosia costulata var. trifoliolata. Sementes das três espécies foram moídas e submetidas à extração salina (NaCl 0,15M -10 %, p/v). Os extratos totais obtidos foram utilizados para quantificar o conteúdo protéico, detectar a atividade residual da tripsina, a atividade hemaglutinante (AHE) e na obtenção do perfil protéico. A atividade residual da tripsina foi observada somente para T. plumbea e S. exasperata, cujos valores foram 4 e 19 %, respectivamente. A AHE foi detectada nos extratos das três espécies, sendo que os extratos totais de T. plumbea e S. exasperata, hemaglutinaram eritrócitos de rato, camundongo e hamster, enquanto que a espécie O. costulata hemaglutinou somente eritrócitos de hamster. O perfil protéico em SDS-PAGE revelou maior ocorrência de proteínas com massa molecular aparente de 10 a 30 kDa para T. plumbea e S. exasperata, enquanto que para O. costulata prevaleceram bandas protéicas com massa molecular variando entre 20-25 kDa. Conclui-se que os extratos totais de O. costulata e S. exasperata, pertencentes à subfamília Papilionoideae, apresentam menor conteúdo de inibidores de tripsina que T. plumbea (Caesalpinioideae) e, quanto à AHE, os resultados mostraram-se diferenciados, mesmo entre as espécies da mesma subfamília, tanto para a concentração mínima hemaglutinante quanto para a especificidade de interação com os eritrócitos.PalavRaS-chavE: Serinoproteinases, Sementes, Lectinas, Eritrócitos.
